Abstract
The present invention is about a pharmaceutical composition, an oral preparation, and an injection preparation containing a G protein-coupled Receptor19 (GPCR19) agonist, specifically sodium taurodeoxycholate (HY2191) and its derivative, as an active ingredient for preventing or treating allergic skin diseases. The present invention is also about an external preparation and a cosmetic composition containing said pharmaceutical composition for preventing and improving allergic skin diseases. The said pharmaceutical composition shows an excellent efficacy in treating and improving allergic dermatitis compared with the steroid ointments or immunosuppressive ointments currently used. The said pharmaceutical composition reduces the level of serum IgE, which is a major factor causing allergic dermatitis, increases TH1 cytokine alleviating allergic dermatitis and reduces TH2 cytokine that exacerbates allergic dermatitis. The said pharmaceutical composition also reduces infiltration of a mast cell, eosinophil, and neutrophil in dermis and ameliorates clinical symptoms such as erythema, hemorrhage, edema, excoriation, erosion, scaling and dryness. Thus, the said pharmaceutical composition can be used to prevent and treat allergic skin diseases.
Core Innovation
The invention provides a GPCR19 agonist composition for preventing or treating allergic skin diseases. The composition includes a G protein-Coupled Receptor 19 (GPCR19) agonist dissolved to be 0.01% to 1% (w/v) in a hyaluronate sodium solvent. The disclosed subject matter is directed to allergic dermatitis, atopic dermatitis, contact dermatitis, hives, and pruritus as allergic skin diseases to be improved.
The disclosed subject matter further describes pharmaceutical preparations and cosmetic compositions containing the GPCR19 agonist. The pharmaceutical preparations include oral, injection, and external preparations, including topical/transdermal patch-type external preparations. The cosmetic care aspect includes a cosmetically effective amount topically applied to a body skin area of a subject in need of care to soothe or alleviate allergic skin diseases.
In the described assessment, the GPCR19 agonist composition is evaluated for improved clinical dermatitis symptoms and modulation of immunological and inflammatory responses. The results described include reduced serum IgE, increased TH1 cytokines (IFN-b3 and IL-2), decreased TH2 cytokine (IL-4), and reduced dermal infiltration of mast cells, eosinophils, and neutrophils. The document also states comparison against vehicle and/or improved effects versus steroid (dexamethasone) or immunosuppressive (tacrolimus) ointments.
Claims Coverage
The document includes two independent claims. The core inventive features focus on the GPCR19 agonist, its dissolution in a hyaluronate sodium solvent at a specified concentration range, and the defined route/form context for prevention or treatment and cosmetic care.
Administering a GPCR19 agonist in hyaluronate sodium solvent
A method of preventing or treating allergic skin diseases by administering an effective amount of a GPCR19 agonist to a subject in need of treatment, wherein the GPCR19 agonist is dissolved so as to be 0.01% to 1% (w/v) in a hyaluronate sodium solvent.
Topically applying a cosmetically effective GPCR19 agonist in hyaluronate sodium solvent
A method for cosmetic care to soothe or alleviate allergic skin diseases by topically applying onto a body skin area of a subject in need of the care a cosmetically effective amount of a GPCR19 agonist, wherein the GPCR19 agonist is dissolved so as to be 0.01% to 1% (w/v) in a hyaluronate sodium solvent.
Across the independent claims, the coverage centers on using a GPCR19 agonist composition dissolved at 0.01% to 1% (w/v) in a hyaluronate sodium solvent, with prevention or treatment achieved by administering an effective amount and cosmetic-care coverage achieved by topical application of a cosmetically effective amount.
