Abstract
Compounds, compositions, and methods comprising a polyamine compound are described, which may be used to kill, disperse, treat, or reduce biofilms, or to inhibit or substantially prevent biofilm formation. In certain aspects, the present invention relates to compounds, compositions, and methods comprising polyamine compounds that have antimicrobial or dispersing activity against a variety of bacterial strains capable of forming biofilms.
Core Innovation
The invention relates to an anti-biofilm composition comprising a carrier and a biocidal polyamine compound, or a salt thereof. The biocidal polyamine compound is defined by a multi-variable polyamine formula with substituent groups designated as R, Rm, R2a, R2b, R3, R4, Y1, Y2, Z1, and Z2, including an alternative where R2a and R2b join to form a spirocycloalkyl ring. The composition comprises a biocidal polyamine compound that comprises at least four primary or secondary amino groups.
The disclosed polyamine compounds are constructed to provide an amphipathic structure by attaching cationic residues, including primary and secondary amino groups, to a lipophilic or hydrophobic backbone and aromatic ring framework options. The document states that dispersing and bactericidal activity are provided through the polyamine cationic residues combined with the lipophilic or hydrophobic moiety, and that increasing primary and secondary amines increases activity.
The structural definitions allow flexibility in substituent selection, including R2a and R2b independently selected from hydrogen, alkyl, and fluoroalkyl, or joined to form a spirocycloalkyl ring. R3 is selected from Z1-Y1-R4 or Z1-Y1-Y2-R4, and R4 is selected from hydrogen, alkyl, fluoroalkyl, alkenyl, alkynyl, arylalkyl, and heteroarylalkyl. The disclosure also includes embodiments as acid salts, including hydrochloride and other acid salts, and representative compound selections shown in figures.
Claims Coverage
The independent claim coverage centers on an anti-biofilm composition with a carrier and a structurally defined biocidal polyamine compound or salt having at least four primary or secondary amino groups, with substituent definitions and optional spirocycloalkyl ring formation. Dependent claims refine the amino-group count, specify particular substituent and salt forms, and narrow the composition to specific product formats and surface/application contexts.
Anti-biofilm composition with carrier and biocidal polyamine compound
An anti-biofilm composition comprising a carrier and a biocidal polyamine compound of a defined formula, or a salt thereof, with substituent groups defined by R, Rm, R2a, R2b, R3, R4, Y1, Y2, Z1, and Z2.
Biocidal polyamine with spirocycloalkyl-forming substituent option
R2a and R2b are independently selected from hydrogen, alkyl, and fluoroalkyl, or alternatively join to form a spirocycloalkyl ring.
At least four primary or secondary amino groups
The biocidal polyamine compound comprises at least four primary or secondary amino groups.
The claim coverage is directed to compositions using a carrier and a defined biocidal polyamine or salt with a spirocycloalkyl-forming substitution option and at least four primary or secondary amino groups, while dependent claims further narrow substituent, salt, and application-format choices.
Stated Advantages
Biocidal polyamines can inhibit and/or disperse biofilms by engaging acidic exopolysaccharide residues within the biofilm extracellular matrix.
The biofilm extracellular matrix limits the efficacy of conventional antibiotics, with reported up to 1000× reduced effectiveness.
Compounds with higher numbers of polyamine chains are described as more effective.
Unexpectedly superior biofilm treatment versus typical antimicrobials.
Biofilm dispersal and rapid antimicrobial killing.
Reduced resistance potential.
Documented Applications
Pipe coating, flush solution, or pipeline flush solution.
Coated on or impregnated into a surface of a medical device.
Biofilm dispersal, biofilm eradication, and/or inhibition on medical device surfaces.
Pipe/pipeline treatment.
Industrial flush solutions.
Sanitizing wipes.
Paints.
Pharmaceutically acceptable carrier-based slow-release formulations.
