Abstract
The present invention provides a bacteriophage with effective antibacterial activity against Staphylococcus strains and in particular MRSA. There is also provided a pharmaceutical composition comprising said bacteriophage and a method of treating a bacterial infection using a composition comprising said bacteriophage.
Core Innovation
The invention relates to Staphylococcus bacteriophage K mutants that comprise one or more mutations within one or more regions corresponding to the nucleotide sequence of wild-type Staphylococcus bacteriophage K (AY176327.1). The specified mutation regions include the region between ORF 18 and ORF 19, the region between ORF 41 and ORF 42, ORF 68, an overlapping region within ORF 86/88/90, ORF 92, ORF 93, ORF 96, and ORF 100.
In particular examples, Staphylococcus bacteriophage K mutants are described with specified point substitutions and a large insertion at position 116111 that is not present in the wild-type sequence. These mutations are located within ORFs including ORF 18/19, ORF 41/42, ORF 68, overlapping ORFs 86/88/90, ORF 92, ORF 93, ORF 96, and ORF 100.
The document explains that the phage mutants broaden antibacterial activity against Staphylococcus strains, including methicillin-resistant Staphylococcus aureus (MRSA). The mutants are associated with pharmaceutical compositions comprising a pharmaceutically acceptable carrier and optionally including an additional phage P68, and the compositions are used for treatment by application to infected surfaces.
Claims Coverage
The independent claim is directed to a Staphylococcus bacteriophage K mutant defined by one or more mutations in specified genome regions, totaling eight selectable regions or ORF areas.
Mutant defined by mutations in selected ORF regions
A Staphylococcus bacteriophage K mutant comprising one or more mutations within one or more regions corresponding to the nucleotide sequence of wild-type Staphylococcus bacteriophage K, selected from the region between ORF 18 and ORF 19; and/or the region between ORF 41 and ORF 42; and/or ORF 68; and/or an overlapping region within ORF 86/88/90; and/or ORF 92; and/or ORF 93; and/or ORF 96; and/or ORF 100.
Inventive coverage is provided by defining Staphylococcus bacteriophage K mutants through one or more mutations located in specific wild-type-corresponding ORF regions. The claim-related material also specifies particular point mutations and a large insertion at position 116111, and connects the mutants to pharmaceutical composition and topical nasal-cavity application.
Stated Advantages
Broaden antibacterial activity against Staphylococcus strains, including methicillin-resistant Staphylococcus aureus (MRSA).
Documented Applications
Pharmaceutical compositions comprising the bacteriophage mutant and a pharmaceutically acceptable carrier are used for treatment by application to infected surfaces.
Topical formulations are described, including topical application to the nasal cavity of a mammal.
