Abstract
The present invention provides antigenic polypeptides expressed during an infection by a pathogenic organism, such as Acinetobacter and compositions comprising these polypeptides. The invention further provides compositions for use in treating, preventing or detecting a bacterial infection, in particular vaccine compositions using the antigenic polypeptides. The invention further provides antibodies directed to said antigenic polypeptides.
Core Innovation
The invention relates to Acinetobacter baumannii infection targets comprising antigenic polypeptides including AB023, AB024, AB025, AB030, AB031, FimA, CsuAB, and OmpA, and nucleic acids encoding the antigenic polypeptides. The invention also relates to vaccine, therapeutic, and diagnostic compositions comprising the antigenic polypeptides and/or nucleic acids, and to antibodies that specifically recognize these targets.
The invention provides nucleic acids encoding the antigenic polypeptides, including genomic DNA and nucleic acids having a specified sequence identity and hybridization under stringent hybridization conditions. The invention further includes expression vectors and host cells capable of producing the encoded antigenic polypeptides.
A key aspect of the invention is an epitope consensus motif PVDFTVAI shown in SEQ ID NO: 36, and antibodies that specifically bind to this epitope consensus motif. The document describes experimental evidence for immunogenicity and detection using convalescent/patient antibody detection by ELISA and immunoblot, and conservation across clinical isolates is highlighted.
Claims Coverage
The provided set includes two independent claims directed to antibodies that specifically bind to the PVDFTVAI epitope consensus motif (SEQ ID NO: 36). Across the independent claims, the core inventive features are specificity for the PVDFTVAI consensus epitope and, for one claim, an IgG format capable of inducing effector function against Acinetobacter baumannii.
Antibody binding to PVDFTVAI epitope consensus motif and IgG effector function
An antibody that specifically binds to the epitope consensus motif PVDFTVAI shown in SEQ ID NO: 36, wherein said antibody is an IgG capable of inducing an effector function towards Acinetobacter baumannii.
Antibody or antigen-binding fragment binding to PVDFTVAI epitope consensus motif
An antibody or antigen-binding fragment thereof that specifically binds to the epitope consensus motif PVDFTVAI (SEQ ID NO: 36).
Overall, the independent claim coverage is directed to antibodies (including antigen-binding fragments) that specifically bind the PVDFTVAI epitope consensus motif (SEQ ID NO: 36), with one independent claim further requiring an IgG format capable of inducing effector function towards Acinetobacter baumannii.
