Abstract

The present invention relates to transgenic green ornamental fish, as well as methods of making such fish by in vitro fertilization techniques. Also disclosed are methods of establishing a population of such transgenic fish and methods of providing them to the ornamental fish industry for the purpose of marketing.

Core Innovation

The invention relates to green transgenic ornamental fish, in particular a transgenic barb comprising a chromosomally integrated expression cassette encoding a green fluorescent protein. The integrated expression cassette is designated as the Green barb 1 transformation event, and this transformation event is comprised in sperm deposited as ECACC accession no. 12103003.

The disclosed subject matter includes fertile transgenic barbs and progeny in which the Green barb 1 transformation event is present. The transgenic barb may be homozygous or heterozygous for the integrated expression cassette, and progeny may exhibit fluorescence and comprise the integrated expression cassette designated as the Green barb 1 transformation event comprised in the sperm deposited as ECACC accession no. 12103003.

The document also describes eggs, sperm, and embryos containing the Green barb 1 transformation event, and production of transgenic barbs using in vitro fertilization, including [procedural detail omitted for safety]. A breeding approach is described in which an obtained fluorescent transgenic barb is bred with a second barb to provide a transgenic barb comprising the Green barb 1 transformation event comprised in the sperm deposited as ECACC accession no. 12103003.

Claims Coverage

The partial claims include two independent claims: a transgenic barb product claim and a method of producing the transgenic barb claim. Across these claims, the coverage centers on chromosomal integration of a green fluorescent protein expression cassette identified as the Green barb 1 transformation event associated with sperm deposited as ECACC accession no. 12103003, and on obtaining and breeding a fluorescent transgenic barb to provide further transgenic barbs comprising the same event.

Chromosomally integrated green fluorescent protein cassette as Green barb 1 transformation event

A transgenic barb comprising a chromosomally integrated expression cassette encoding a green fluorescent protein, wherein the integrated expression cassette is designated as the Green barb 1 transformation event comprised in the sperm deposited as ECACC accession no. 12103003.

Producing transgenic barb by breeding a fluorescent Green barb 1 transgenic barb with a second barb

A method of producing a transgenic barb comprising obtaining a transgenic barb that exhibits fluorescence and comprises a chromosomally integrated expression cassette encoding a green fluorescent protein, wherein the integrated expression cassette is designated as the Green barb 1 transformation event comprised in the sperm deposited as ECACC accession no. 12103003; and breeding the obtained transgenic barb with a second barb to provide a transgenic barb comprising the Green barb 1 transformation event comprised in the sperm deposited as ECACC accession no. 12103003.

Overall, the claim set establishes protection around the specific Green barb 1 transformation event defined by a chromosomally integrated green fluorescent protein expression cassette associated with the ECACC-deposited sperm, including transgenic barb products and methods that obtain a fluorescent Green barb 1 transgenic barb and breed it with a second barb to maintain the same transformation event.
