Abstract

A sample acquiring device, an apparatus and a method for spectrophotometric measurement of high density lipoprotein (HDL) and at least one of total cholesterol (TC), triglycerides (TG) and glucose (FPG).

Core Innovation

The invention describes a sample acquiring device that includes at least one inlet for receiving a body fluid sample and at least one capillary sample reception cavity in capillary communication with the inlet. A first pathway and a second pathway are each in communication with the inlet and a separate measurement cavity, and the first pathway and the second pathway are in communication with the capillary sample reception cavity such that spontaneous flow from the capillary sample reception cavity is prevented. The device forces the body fluid sample from the capillary sample reception cavity into a first centrifugation cavity of the first pathway and a first centrifugation cavity of the second pathway by subjecting the sample acquiring device to a first centrifugation.

The first centrifugation cavities and the second centrifugation cavities have a lower portion and an upper portion oriented relative to each other such that centrifugation forces sample toward the lower portion. Each first capillary cavity is in capillary communication only with the upper portion of its corresponding centrifugation cavity, such that transfer of at least a fraction of the body fluid sample occurs from the centrifugation cavities to the capillary cavities. The device further prevents spontaneous flow from the first capillary cavities by subjecting the sample acquiring device to a second centrifugation that forces fractions of the body fluid sample into second centrifugation cavities, and then to second capillary cavities via further capillary communication through upper portions.

The device provides that spontaneous flow from the second capillary cavities is prevented and that fractions of the body fluid sample in the second capillary cavities are forcible into the first measurement cavity and the second measurement cavity by subjecting the sample acquiring device to a third centrifugation. At least one chromophore is provided in at least one selected location of the second pathway and similarly in the first pathway depending on the pathway. A precipitation agent is provided in the first capillary cavity of the second pathway, and the device is arranged so that each pathway separately delivers sample fractions for measurement.

Claims Coverage

The partial content provides one independent claim. It defines a two-path sample acquiring device with sequential centrifugation and capillary-to-centrifugation-to-capillary transfers that prevent spontaneous flow and deliver separate measurement cavities, including a precipitation agent in the second pathway and chromophore placement for measurement.

Two-path capillary/centrifugation architecture with separate measurement cavities

A sample acquiring device with a first pathway and a second pathway in capillary communication with a capillary sample reception cavity, where spontaneous flow from the capillary sample reception cavity is prevented and body fluid sample is forcibly moved into separate centrifugation cavities by a first centrifugation and then toward separate measurement cavities.

Upper-portion-only capillary communication after centrifugation

Centrifugation cavities have lower and upper portions such that centrifugation forces sample toward the lower portion, and each capillary cavity is in capillary communication only with the upper portion of its corresponding centrifugation cavity to enable transfer of at least a fraction into the capillary cavities.

Sequential centrifugations to transfer fractions between cavities while preventing spontaneous flow

The device prevents spontaneous flow from the first capillary cavities and forces fractions from the first capillary cavities into second centrifugation cavities by a second centrifugation, and then forces fractions from the second capillary cavities into the first and second measurement cavities by a third centrifugation.

Precipitation agent in a capillary cavity of the second pathway

A precipitation agent is provided in the first capillary cavity of the second pathway.

Chromophore placement for measurement in pathway cavities and measurement cavities

At least one chromophore is provided in selected locations of the first pathway and the second pathway, including the second capillary cavity, the transfer cavity, or the first or second measurement cavity, depending on the pathway.

Across the provided independent claim, the main inventive concept is a device that uses separate first and second pathways with sequential centrifugations and upper-portion-only capillary communication to prevent spontaneous flow and to deliver separately measured sample fractions, with a precipitation agent in the second pathway and chromophore(s) provided for measurement within designated cavities.
