Abstract

A composition prepared from a cranberry hull enzyme-treated composition which reduces or inhibits the adhesion of microorganisms to cells having α-Gal-(1-4)-Gal terminal oligosaccharide receptors for adhesion.

Core Innovation

The invention relates to a novel xyloglucan oligosaccharide composition, designated anti-microbial adhesion inhibitory fraction A6, derived from enzyme treated cranberry hulls of Vaccinium macrocarpon. This composition reduces or inhibits the adhesion of microorganisms to cells possessing α-Gal-(1-4)-Gal terminal oligosaccharide receptors, thereby potentially preventing bacterial colonization and infection.

The composition comprises an acetylated, neutral-sugar-rich polysaccharide having a weight-average molar mass of approximately 10.2×10³±2 Da, a Z-average hydrodynamic radius of about 2.0±0.2 nm, and a weight-average intrinsic viscosity of roughly 0.048±0.001 dL/g. The monosaccharide composition of the active fraction A6 is dominated by glucose, arabinose, and xylose, indicating it is a hemicellulose, specifically an arabino-xyloglucan with a predominant SSGG block sequence not previously characterized in the Ericales.

The problem being addressed is the frequent misuse of antibiotics leading to antibiotic-resistant bacterial strains and recurrent infections such as urinary tract infections caused primarily by E. coli. There remains a need for compositions and methods that reduce bacterial adhesion to host cells, especially human and mammalian cells, to prevent bacterial colonization and infection without reliance on antibiotics. The invention provides a composition and method to address this by inhibiting bacterial adhesion to α-Gal-(1-4)-Gal terminal oligosaccharide receptors on host cells.

Claims Coverage

The patent includes three independent claims covering compositions and methods for inhibiting bacterial adhesion using cranberry-derived oligosaccharide fractions. The inventive features focus on the composition, its method of preparation, and its application for reducing bacterial adhesion.

Anti-microbial composition comprising enzyme-treated cranberry hull fractions

An anti-microbial composition in tablet or capsule form containing an effective amount of an anti-microbial adhesion inhibitory composition prepared by: (a) providing cranberry hulls; (b) depectinizing the hulls using pectinase enzyme treatment to form an enzyme treated composition; (c) removing debris by decantation or centrifugation to obtain a first liquid; (d) spray drying to form composition A1; (e) fractionating A1 via C18 Flash Chromatography to produce composition A2; (f) freeze drying A2; (g) purifying A2 with LH20 gel column chromatography to obtain the adhesion inhibitory fraction A6; (h) freeze drying A6 to crystalline powder; (i) mixing with a carrier to provide the final anti-microbial composition.

Anti-microbial composition comprising arabino-xyloglucan with SSG structure

An anti-microbial composition prepared as above, wherein the active composition comprises arabino-xyloglucan having an SSG structure, conferring anti-adhesion properties against bacteria binding to α-Gal-(1-4)-β-Gal terminal oligosaccharide receptors.

Method for reducing bacterial adhesion using cranberry-derived composition

A method for reducing the adhesion of bacteria to cells having α-Gal-(1-4)-Gal terminal oligosaccharide receptors by administering an effective amount of the composition prepared from enzyme-treated cranberry hull fractions comprising the anti-microbial adhesion inhibitory fraction A6.

The claims encompass compositions containing the cranberry-derived anti-microbial adhesion inhibitory fraction A6 prepared by a defined enzymatic and chromatographic process, the specific arabino-xyloglucan structure with anti-adhesion activity, and methods of reducing bacterial adhesion through administration of such compositions.

Stated Advantages

The anti-microbial fraction A6 effectively reduces or inhibits bacterial adhesion to human and mammalian cells expressing α-Gal-(1-4)-Gal oligosaccharide receptors, addressing bacterial colonization without antibiotics.

The composition utilizes a natural cranberry-derived polysaccharide, providing an alternative to antibiotic treatments and potentially reducing antibiotic resistance and recurrent infections.

The active component exhibits specific blocking activity against uropathogenic and verotoxigenic E. coli strains, relevant to urinary tract infection prevention.

Documented Applications

Reducing adhesion of pathogenic bacteria such as E. coli to human bladder epithelial cells to prevent urinary tract infections.

Reducing adhesion of verotoxigenic E. coli to human colonic epithelial cells to prevent infection.

Use as a medicament, nutritional composition, or pharmaceutical composition in tablet or capsule form for oral administration.
