Abstract

The present invention is directed to the field of phage therapy for the treatment and control of bacterial infections. In particular, the present invention is directed to the novel bacteriophage F387/08, F391/08, F394/08, F488/08, F510/08, F44/10, and F125/10, isolated polypeptides thereof, compositions comprising one or more of the novel bacteriophage and/or isolated polypeptides, as well as to methods for the treatment and prevention of bacterial infections using same, either alone or in combination with other antibacterial therapies, e.g., antibiotics and/or other phage therapies.

Core Innovation

The invention relates to phage therapy directed to bacterial infections, including treating or reducing the incidence of a Pseudomonas aeruginosa bacterial infection in a subject in need thereof. The approach is based on administering a pharmaceutical composition that includes a pharmaceutically acceptable carrier and a purified bacteriophage having a nucleic acid sequence of SEQ ID NO:4.

The disclosed subject matter links specific bacteriophages and phage-derived polypeptides to antibacterial activity against nosocomial pathogens. It describes bacteriophages including F387/08, F391/08, F394/08, F488/08, F510/08, F44/10, and F125/10, and also describes antibacterial phage-derived polypeptides, including lysins/endolysins, CHAP amidase domains, and tail proteins.

The disclosure additionally addresses use in pharmaceutical compositions and delivery contexts, including phage cocktails and administration contexts. It further describes diagnostic use based on susceptibility to phage or polypeptide activity, and concepts involving biosensor and tail-protein recognition, while also covering fragments, variants, or derivatives of the described entities using amino acid sequence identity thresholds.

Claims Coverage

The document includes one independent claim covering a treatment method for reducing the incidence of a Pseudomonas aeruginosa bacterial infection. The inventive features in the independent claim focus on the specific bacteriophage defined by the nucleic acid sequence of SEQ ID NO:4 and the formulation as a pharmaceutical composition with a pharmaceutically acceptable carrier.

Treatment of Pseudomonas aeruginosa infection with SEQ ID NO:4 phage composition

A method of treating or reducing the incidence of a Pseudomonas aeruginosa bacterial infection in a subject in need thereof by administering an effective amount of a pharmaceutical composition comprising a pharmaceutically acceptable carrier and a purified bacteriophage having a nucleic acid sequence of SEQ ID NO:4.

Across the independent claim coverage provided, the core inventive concept is the use of a purified bacteriophage defined by the nucleic acid sequence SEQ ID NO:4 in a pharmaceutically acceptable carrier formulation for treating or reducing the incidence of Pseudomonas aeruginosa infection.
