Abstract

The invention proposes a cell-free graft, comprising (i) a cohesive, structure-forming matrix with open porosity made from a biologically and pharmaceutically acceptable material and (ii) serum. In one particularly preferred embodiment, the matrix additionally contains a gel. By virtue of a second aspect, a method of producing such a cell-free graft is proposed, whereby the matrix and the gel, if one is provided, is placed in contact with the serum. The graft may optionally be dried with serum. Alternatively, the matrix and the gel, if one is provided, are in the dried state before being placed in contact with the serum.By virtue of a third aspect, finally, the invention proposes the use of the cell-free graft for regenerating tissue and in particular for regenerating cartilage and/or bone.

Core Innovation

The invention relates to a cell-free graft for cartilage regeneration and/or bone regeneration. The graft includes a cohesive, structure-forming matrix with open porosity made from a biologically and pharmaceutically acceptable material, selected from fleece or felt structures, wadding, wool, and any combinations thereof. The graft further includes human serum in a concentration in the range of about 0.01% to 50% by volume.

In addition to the cohesive open-porosity matrix and human serum, the graft includes a gel applied to at least one side of the matrix and at least partially penetrating the matrix. The gel is selected from polysaccharides, hyaluronic acid, alginate, agarose and chitosan, as well as mixtures thereof. The disclosed formulations are described as capable of recruiting mesenchymal precursor cells.

The disclosure addresses improved recruitment efficiency of mesenchymal precursor cells. The invention uses human serum in the graft matrix, and optionally a gel that acts as a barrier to improve cell selectivity under joint stresses. Experimental examples show in vitro chemotactic recruitment by human serum across formulations versus specific growth factors/chemokines.

Claims Coverage

The independent claim coverage centers on a cell-free graft defined by three structural components: a cohesive open-porosity matrix from fleece/felt-type materials, human serum within a specified concentration range, and a gel applied to at least one matrix side that at least partially penetrates the matrix.

Cohesive open-porosity structure-forming matrix from fleece or felt structures

A cohesive, structure-forming matrix with open porosity made from a biologically and pharmaceutically acceptable material selected from fleece or felt structures, wadding, wool, and any combinations thereof.

Human serum in a defined concentration range

Human serum in a concentration in the range of about 0.01% to 50% by volume.

Gel applied to a matrix side and partially penetrating selected polysaccharides

A gel applied to at least one side of the matrix and at least partially penetrates it, selected from polysaccharides, hyaluronic acid, alginate, agarose and chitosan, as well as mixtures thereof.

The claim set focuses on a cell-free graft that combines a cohesive open-porosity fleece/felt-type matrix with human serum at about 0.01% to 50% by volume, and a penetrating gel layer on at least one matrix side selected from polysaccharides including hyaluronic acid, alginate, agarose, and chitosan.

Stated Advantages

Increases recruitment efficiency of mesenchymal precursor cells compared to growth factors/chemokines.

Enables storage/handling simplification compared to growth factors/chemokines.

Improves cell selectivity under joint stresses through use of a barrier gel.

Documented Applications
