Abstract
A reagent or particles having reagent supported thereon are applied to a matrix material by a contact printing process for example in which the matrix material is brought into contact with a rotating contact roll having the reagent or particles having reagent supported thereon disposed thereon so as to transfer the reagent or particles having reagent supported thereon to the matrix material. Such a contact printing processes allow high speed and high volume production of an assay device.
Core Innovation
A test device for wiping over a surface to extract a sample includes two impermeable layers with an absorbent matrix material arranged between the impermeable layers and located immediately adjacent both impermeable layers. The matrix material carries a reagent and has a patterned surface, and one impermeable layer has a plurality of openings aligned with the matrix material through which the sample is absorbed into the matrix material as facilitated by the wiping over the surface.
A further test device uses two impermeable layers with an absorbent matrix material arranged between them immediately adjacent the impermeable layers, with the matrix material having a reagent applied thereto. One impermeable layer has at least one opening aligned with the matrix material through which the sample is absorbed into the matrix material as facilitated by the wiping over the surface, and a one-way flow semi-permeable layer extends across the opening and is located immediately adjacent the absorbent matrix material and a second of the two impermeable layers.
The device architecture can also be configured with two flexible impermeable layers and a flexible layer of absorbent matrix material arranged between them, where the matrix material carries a reagent and has a patterned surface and one flexible impermeable layer has at least one opening aligned with the flexible absorbent matrix material. A one-way flow flexible semi-permeable layer extends across the opening and is located immediately adjacent the flexible absorbent matrix material and between the two flexible impermeable layers, being made of a semi-permeable material that allows the sample to pass towards the flexible absorbent matrix material while limiting backflow of the reagent.
In additional device structures, the absorbent matrix material carries a culture medium and either a conductive material or a thin film battery for heating or warming the culture medium, combined with a reagent applied to the matrix. The test device still includes two impermeable layers with an opening in one impermeable layer aligned with the matrix material so that the sample may be absorbed into the matrix material as facilitated by wiping.
Claims Coverage
The independent claims cover wiping-over sample extraction test devices comprising a layered structure, sample entry via aligned openings, and enhanced localization of sample and/or reaction through patterned absorbent matrix surfaces and/or a one-way flow semi-permeable layer that limits reagent backflow; additional independent claims further add a culture medium and heating or warming via a conductive material or a thin film battery.
Patterned absorbent matrix surface with aligned openings for reduced spreading and increased sample concentration
A test device for wiping over a surface to extract a sample comprising two impermeable layers; an absorbent matrix material arranged between the impermeable layers and located immediately adjacent both impermeable layers, the matrix material carrying a reagent and having a patterned surface; and one impermeable layer having a plurality of openings aligned with the matrix material through which the sample may be absorbed into the matrix material as facilitated by the wiping over the surface, wherein the patterned surface of the matrix material reduces spreading and increases a concentration of the sample in regions corresponding to the plurality of openings.
One-way flow semi-permeable layer limiting reagent backflow through aligned openings
A test device for wiping over a surface to extract a sample comprising two impermeable layers; an absorbent matrix material arranged between the impermeable layers located immediately adjacent a first of the two impermeable layers, the matrix material having a reagent applied thereto, one impermeable layer having at least one opening aligned with the matrix material through which the sample may be absorbed into the matrix material as facilitated by the wiping over the surface; and a one-way flow semi-permeable layer extending across the at least one opening and located immediately adjacent the absorbent matrix material and a second of the two impermeable layers, the one-way flow semi-permeable layer being made of a semi-permeable material which only allows the sample to pass in a direction towards the matrix material, but limits backflow of the reagent from the matrix material.
Flexible patterned absorbent matrix with one-way flow flexible semi-permeable layer
A test device for wiping over a surface to extract a sample comprising two flexible impermeable layers; a flexible layer of absorbent matrix material arranged between the flexible impermeable layers, the matrix material having a reagent applied thereto and having a patterned surface; one flexible impermeable layer having at least one opening aligned with the flexible absorbent matrix material and through which the sample may be absorbed into the flexible absorbent matrix material as facilitated by the wiping over the surface; and a one-way flow flexible semi-permeable layer extending across the at least one opening and located immediately adjacent the flexible absorbent matrix material and between the two flexible impermeable layers, the one-way flow flexible semi-permeable layer being made of a semi-permeable material which only allows the sample to pass in a direction towards the flexible absorbent matrix material, but limits backflow of the reagent from the flexible absorbent matrix material, wherein the patterned surface of the flexible absorbent matrix material reduces spreading and increases a concentration of the sample in regions corresponding to the at least one opening.
Culture medium heating or warming using a conductive material or thin film battery
A test device for wiping over a surface to extract a sample comprising two impermeable layers; a layer of absorbent matrix material arranged between the impermeable layers and located immediately adjacent both impermeable layers, the matrix material carrying a reagent, a culture medium, and either a conductive material or a thin film battery for heating or warming the culture medium; and one of the impermeable layers having a plurality of openings aligned with the matrix material and through which the sample may be absorbed into the matrix material as facilitated by the wiping over the surface.
Culture medium heating or warming combined with semi-permeable layer limiting reagent backflow
A test device for wiping over a surface to extract a sample comprising two impermeable layers; an absorbent matrix material arranged between the impermeable layers and located immediately adjacent a first of the two impermeable layers, the matrix material having a reagent applied thereto and carrying a culture medium and either a conductive material or a thin film battery for heating or warming the culture medium, one of the impermeable layers having at least one opening aligned with the matrix material and through which the sample may be absorbed into the matrix material as facilitated by the wiping over the surface; and a semi-permeable layer extending across the at least one opening and located immediately adjacent the absorbent matrix material and a second of the two impermeable layers, the semi-permeable layer being made of a semi-permeable material which allows the sample to pass therethrough whilst limiting backflow of the reagent from the matrix material.
Across the independent claims, the central coverage is a wiping test device that extracts a sample into an absorbent matrix using aligned openings through impermeable layers, with either patterned matrix surfaces to reduce spreading and increase sample concentration, a one-way flow semi-permeable layer to limit reagent backflow, or both. Additional independent claim coverage includes adding a culture medium and heating or warming it using either a conductive material or a thin film battery, optionally alongside a semi-permeable layer that limits backflow.
