Abstract

A Product is described and which contains at least one type of ligand bound to a separation material and which allows selective binding or cleavage of a biomolecule for example in human blood.

Core Innovation

The invention relates to a separation-material product comprising a ligand immobilized on a matrix for selective binding and/or cleavage of biomolecules. The disclosed ligand includes saccharides, including blood-group determinants, bound directly or immobilized via optional spacer chemistries to the matrix.

The matrix is described as a separation matrix such as agarose or cross-linked agarose, including forms that enable penetration of IgM/IgG. The disclosure further includes spacer-linked saccharide-matrix structures and examples of spacer/linker chemistries used to immobilize the blood-group determinants.

The disclosure also describes combined or connected extracorporeal uses with immobilized proteolytic and/or hydrolytic enzymes for cleavage of antibodies. In particular, the matrix further comprises proteolytic or hydrolytic enzymes with specificity to cleave IgG bound to the matrix, with IdeS and optional EndoS mentioned, and the overall system is used with blood or plasma to reduce antibodies, including anti-A and anti-B, and to cleave IgG and/or IgM during blood donation or ABO-incompatible transplantation.

Claims Coverage

The patent defines one independent combined product claim with two principal inventive features: a column-matrix carrying blood group A and blood group B saccharides for binding anti-A and anti-B antibodies, and an immobilized proteolytic or hydrolytic enzyme on the same matrix for cleavage of IgG bound to the matrix.

Column-matrix with both anti-A and anti-B saccharide binding specificities

A column and a matrix filled into the column, wherein at least one blood group A saccharide with specificity to bind to anti-A antibodies and at least one blood group B saccharide with specificity to bind anti-B antibodies is bound to the matrix, optionally via a spacer.

Matrix further comprising an IgG-cleaving proteolytic or hydrolytic enzyme

The matrix further comprising at least one proteolytic or hydrolytic enzyme with specificity to cleave IgG bound to the matrix.

Overall, the claim coverage centers on a combined column-matrix product that co-localizes blood-group saccharide binding for anti-A and anti-B antibodies with an immobilized proteolytic or hydrolytic enzyme that cleaves IgG bound to the matrix.

Stated Advantages

Enables selective binding and cleavage of biomolecules in human blood or plasma using the combined product.

Supports reduction of antibodies, including anti-A antibodies and anti-B antibodies, during extracorporeal blood/plasma treatment.

Provides cleavage of IgG bound to the matrix; additionally, the disclosure context includes cleavage of IgM.

Documented Applications

Extracorporeal treatment that passes human blood or plasma through the combined product to extracorporeally remove anti-A and/or anti-B antibodies and/or cleave IgG and/or IgM during blood donation.

Extracorporeal treatment for ABO-incompatible transplantation of a sensitized patient, including removal of anti-A and/or anti-B antibodies and cleavage of IgG and/or IgM.
