Abstract

The present invention provides a novel glyceroglycolipid produced by Mycoplasma pneumoniae. The glyceroglycolipid can be used as a diagnostic marker for a disease caused by Mycoplasma pneumoniae.

Core Innovation

The invention relates to a glyceroglycolipid antigen associated with Mycoplasma pneumoniae, including a defined glycerol/diacylglycerol backbone bearing a β-linked disaccharide of Galβ-6Galβ-3DAG or Glcβ-6Galβ-3DAG. The antigen is described as having variants defined by the R1/R2 pattern and acyl chain variants corresponding to specified R3 substituent forms, and includes acid/base salts.

The invention further provides diagnostic compositions and diagnostic agents, including diagnostic kits, that use the above-described Mycoplasma pneumoniae glyceroglycolipid antigen for immunological detection. The described detection includes antibody-based immunodetection to detect M. pneumoniae infection and related diseases.

Supporting evidence described in the partial content includes TLC-immunostaining using Guillain-Barre patient sera and ELISA results using Glcβ-6Galβ-3DAG, including ROC curve evaluation and discussion of antibody reactivity and cross-reactivity, as well as a test-strip concept for immunochromatography.

Claims Coverage

The relevant independent claims cover isolated and synthesized glyceroglycolipid-type compounds defined by a constrained R1/R2 OH/H substitution rule and defined R3 alkyl-chain options, including salts, and a diagnostic kit for detecting a disease caused by Mycoplasma pneumoniae.

Isolated compound defined by a general formula with R1/R2 OH/H pattern and defined R3 alkyl-chain options

An isolated compound represented by a general formula wherein when R1=OH then R2=H and when R1=H then R2=OH, and at least one of R3 is —(CH2)14CH3, with the other R3 being —(CH2)14CH3 or —(CH2)16CH3, or a salt thereof.

Diagnostic kit for detecting disease caused by Mycoplasma pneumoniae using the isolated compound

A diagnostic kit including the compound of the isolated-compound general formula and manufacturer’s instructions, for detecting a disease caused by Mycoplasma pneumoniae.

Isolated compound as an isolated natural compound derived from Mycoplasma pneumoniae

An isolated compound represented by the same general formula wherein when R1=OH then R2=H and when R1=H then R2=OH, and at least one of R3 is —(CH2)14CH3, with the other R3 being —(CH2)14CH3 or —(CH2)16CH3, or a salt thereof, wherein the compound is an isolated natural compound derived from Mycoplasma pneumoniae.

Chemically synthesized compound defined by a general formula with R1/R2 OH/H pattern and defined R3 alkyl-chain options

A chemically synthesized compound represented by a general formula wherein when R1=OH then R2=H and when R1=H then R2=OH, and at least one of R3 is —(CH2)14CH3, with the other R3 being —(CH2)14CH3 or —(CH2)16CH3, or a salt thereof.

Overall, the claim set centers on isolatable and/or synthesized glyceroglycolipid-type compounds defined by the R1/R2 OH/H substitution rule and defined R3 alkyl-chain options, including salts, and includes a diagnostic kit use for detecting a Mycoplasma pneumoniae-caused disease.
