Abstract

Bis-(4,4′dimethyl-2,2′bipyridyl) picolinate osmium complexes are useful as mediators in the electrochemical test strips, such as those used in the detection of glucose.

Core Innovation

The disclosed invention relates to electrochemical glucose biosensors that use a dual electron-transfer system. A redox mediator rapidly regenerates glucose oxidase, while a separate high-solubility redox shuttle provides most electrode current and reacts slowly with oxygen. This arrangement addresses oxygen-related interference in glucose detection by separating roles between fast enzyme regeneration and slower electrode current contribution.

Selection and tuning are described using the relative redox potentials (E0) of the mediator and the shuttle, the shuttle oxygen reactivity, and solubility together with a concentration ratio. The applied potential (Vapp) is tuned so that reactions proceed in a controlled manner, enabling the sensor to maintain the dual electron-transfer behavior rather than relying on a single electron-transfer component.

Examples are described for mediator and shuttle combinations, including ferricyanide as a shuttle and osmium(II/III) redox mediator compounds. The mediator compounds include osmium complexes based on bipyridyl picolinate/picolinate-like ligands, including PF6 salts, and performance is compared for dual versus single mediator configurations. A sensor structure is described that includes a support and sample chamber, electrodes, and a reagent composition containing glucose oxidase, the mediator compound, and the shuttle compound.

Claims Coverage

The partial content provides two independent claims: one directed to a specific redox mediator compound formula and one directed to a disposable glucose test strip in which the redox mediator is defined by that formula. Together, the claim set specifies the chemical definition of the mediator (n and counterions Y) and its use as the redox mediator component for regenerating enzyme in a disposable glucose measurement strip.

Redox mediator compound formula defined by n and counterions Y

A compound of the formula wherein n is 1 or 2, and Y is one or more counterions with a charge equal and opposite to n.

Disposable glucose test strip with a redox mediator of the defined formula

A disposable test strip for measurement of glucose in a blood sample containing an enzyme that reacts with glucose and a redox mediator compound for regenerating enzyme, wherein the redox mediator comprises a compound of the formula wherein n is 1 or 2, and Y is one or more counterions with a charge equal and opposite to n.

Across the independent claims, the core coverage is the defined redox mediator compound formula (n limited to 1 or 2 with counterions Y having the opposite charge) and its incorporation as the redox mediator in a disposable blood glucose test strip for regenerating the glucose-reacting enzyme.

Stated Advantages

Provides electrochemical glucose detection using a dual electron-transfer system where the mediator rapidly regenerates glucose oxidase and the shuttle provides most electrode current while reacting slowly with oxygen.

Controls oxygen-related interference by separating mediator function from shuttle oxygen reactivity.

Documented Applications
