Abstract

The present invention is directed to isolated bacteriophage having strong lytic activity against strains of Clostridium perfringens, and methods of using that bacteriophage, and/or progeny or derivatives derived therefrom, to control the growth of Clostridium perfringens in various settings.

Core Innovation

The disclosure describes isolated bacteriophage strains CPAS-16 and CPLV-42 deposited under ATCC accession numbers PTA-8481 and PTA-8483, respectively. These isolated bacteriophage strains have lytic activity against Clostridium perfringens strains and variants thereof, and variants have the same phenotypic characteristics and the same lytic activity as the bacteriophage strain.

The disclosure describes compositions and preparations comprising the isolated bacteriophage strains. It also relates to lysing targeted C. perfringens while not lysing tested non-C. perfringens strains, and to detection and diagnostics of viable C. perfringens using reporter/reaction readouts, including ATP/adenosine kinase release and luminescence, and epidemiological typing.

The disclosure describes documented characterization including RFLP profiles using Restriction Fragment Length Polymorphism, and it presents lytic specificity across multiple C. perfringens strains and non-target species. The disclosure also includes examples and related assays based on phage-triggered lysis products and phage progeny, with related criteria for variants.

Claims Coverage

The independent claim(s) present one core inventive subject matter: isolated bacteriophage strains CPAS-16 or CPLV-42 deposited under specific ATCC accession numbers, having lytic activity against Clostridium perfringens strains, including variants retaining specified phenotypic and lytic characteristics.

Isolated bacteriophage strain CPAS-16 or CPLV-42 deposited under ATCC accession numbers

An isolated bacteriophage strain CPAS-16 or CPLV-42 deposited under ATCC accession number PTA-8481 or PTA-8483 respectively, having lytic activity against Clostridium perfringens strains, and variants thereof.

Variants retaining the same phenotypic and lytic activity

Variants have the same phenotypic characteristics of said bacteriophage strain and the same lytic activity of said bacteriophage strain against Clostridium perfringens strains.

Across the independent claim, the inventive concept is the specific isolated bacteriophage strains CPAS-16 (ATCC PTA-8481) and CPLV-42 (ATCC PTA-8483) and variants that retain the same phenotypic characteristics and the same lytic activity against Clostridium perfringens strains.

Stated Advantages

Lysis of targeted C. perfringens while not lysing tested non-C. perfringens strains.

Detection/diagnostics of viable C. perfringens using reporter/reaction readouts such as ATP/AK release and luminescence.

Epidemiological typing.

Documented Applications

Food preservation/food safety, including use on foods, processing equipment, surfaces, and packaging materials.

Environmental control, including biofilm prevention and facility decontamination.

Decontamination of medical, veterinary, and animal-husbandry environments.

Prevention or treatment of animal and human diseases, including necrotic enteritis in chickens.

Detection/diagnostics of viable C. perfringens, including reporter/reaction readouts and epidemiological typing.
