Abstract
The present invention is directed to isolated bacteriophage having strong lytic activity against strains of Clostridium perfringens, and methods of using that bacteriophage, and/or progeny or derivatives derived therefrom, to control the growth of Clostridium perfringens in various settings.
Core Innovation
The invention relates to isolated bacteriophage strain CPAS-7, CPAS-12, or CPAS-15 deposited under specific ATCC accession numbers, each having lytic activity against Clostridium perfringens strains. The bacteriophages and variants are characterized by the same phenotypic characteristics as the deposited bacteriophage strain and the same lytic activity against Clostridium perfringens strains.
The document further discloses that the bacteriophages can be used to control colonization and/or growth of Clostridium perfringens in food, food processing equipment, animal environments, and medical/veterinary environments. It also states prevention and/or treatment of C. perfringens diseases in animals and/or humans, as well as environmental decontamination and prevention of biofilms.
Beyond lytic control, the document describes diagnostic detection of viable C. perfringens using phage-triggered ATP/adenosine kinase release and luciferase/luciferin readouts, including a luciferase reporter phage. The document also describes epidemiological typing and characterization based on RFLP profiling, including variants with substantially equivalent RFLP profile.
Claims Coverage
The partial content identifies one independent claim focused on an isolated bacteriophage strain CPAS-7, CPAS-12, or CPAS-15 deposited under specific ATCC accession numbers and retaining lytic activity against Clostridium perfringens strains. Variants with the same phenotypic characteristics and same lytic activity are also covered, and dependent claims refine the scope toward isolated progeny and compositions comprising the bacteriophage or progeny.
Deposited isolated bacteriophage with lytic activity against Clostridium perfringens strains
An isolated bacteriophage strain CPAS-7, CPAS-12, or CPAS-15 deposited under ATCC accession number PTA-8478, PTA-8479 or PTA-8480 respectively, having lytic activity against Clostridium perfringens strains and variants thereof.
Variants retaining phenotypic characteristics and lytic activity
Variants thereof wherein said variants have the same phenotypic characteristics of said bacteriophage strain and the same lytic activity of said bacteriophage strain against Clostridium perfringens strains.
Isolated progeny preserving the same phenotype and lytic activity
An isolated progeny of the bacteriophage strain retaining the same phenotypic characteristics and the same lytic activity against Clostridium perfringens strains.
Composition comprising the isolated bacteriophage strain
A composition that includes the isolated bacteriophage strain described in claim 1.
Composition comprising the isolated progeny
A composition that includes the isolated bacteriophage strain specified in claim 2.
Overall claim coverage centers on deposited isolated bacteriophage strains CPAS-7, CPAS-12, and CPAS-15 with lytic activity against Clostridium perfringens strains, including variants and isolated progeny that retain the same phenotypic characteristics and the same lytic activity, as well as compositions comprising the bacteriophage or progeny.
