Abstract
The present disclosure relates to bacteriophages and compositions capable of infecting and killing Staphylococcus, and use of the same for treating Staphylococcus, e.g. Staphylococcus aureus, bacterial infections.
Core Innovation
The invention describes Staphylococcus aureus-targeting bacteriophages, including ARSA0001 and ARSA0002, and therapeutic compositions for treating Staphylococcus aureus infections. The claims cover compositions containing two or more bacteriophages, at least one having a polynucleotide sequence with at least 97% identity to SEQ ID NO:2, as well as compositions containing one or more bacteriophages meeting that sequence-identity feature.
The disclosed phages retain infectivity after plasma exposure, reduce biofilm biomass, and show additive to synergistic activity with vancomycin, including against vancomycin-resistant Staphylococcus aureus. The disclosure identifies treatment of Staphylococcus aureus infections, including infections involving resistant strains, as the therapeutic application without explicitly stating the specific background problem.
Claims Coverage
The two independent claims cover methods of treating a Staphylococcus aureus infection using therapeutic compositions that contain bacteriophages with a specified sequence-identity relationship to SEQ ID NO:2. They define two inventive features.
Treatment with two or more bacteriophages
The first independent claim covers administering a therapeutic composition to a subject in need of treatment, where the composition comprises two or more bacteriophages and at least one bacteriophage comprises a polynucleotide sequence with at least 97% identity to SEQ ID NO:2.
Treatment with one or more bacteriophages
The second independent claim covers administering a therapeutic composition to a subject in need of treatment, where the composition comprises one or more bacteriophages, including a bacteriophage with a polynucleotide sequence with at least 97% identity to SEQ ID NO:2.
Together, the independent claims cover treatment of Staphylococcus aureus infection using compositions with one or more bacteriophages meeting the stated sequence-identity feature; one claim specifically requires two or more bacteriophages.
Stated Advantages
The lead phages retained about 94% infectivity after plasma exposure, while the tested comparators were substantially inactivated.
AP-SA02 eradicated approximately 60–90% of biofilm biomass.
AP-SA02 showed additive to synergistic activity with vancomycin, including against vancomycin-resistant Staphylococcus aureus.
Documented Applications
Treatment of Staphylococcus aureus infections, including infections involving methicillin-resistant, vancomycin-resistant, and vancomycin-intermediate Staphylococcus aureus.
