Abstract
Disclosed herein are topical formulations of cannabidiol, such as nanoparticulate cannabidiol, methods of making such compositions, and their use in treating autoimmune diseases and disorders, e.g., alopecia areata.
Core Innovation
The invention relates to topical cannabidiol (CBD) formulations that use nanoparticulate cannabidiol to achieve skin penetration and immunomodulation for autoimmune diseases. The patent focuses on topical delivery with nanoparticulate CBD, including substantially pure cannabidiol, and emphasizes penetration into the skin in terms of epidermis and dermis capability.
The compositions include nanoparticulate cannabidiol having a defined cannabidiol content and an average particle size range. The nanoparticulate cannabidiol is capable of penetrating into the skin and is described as substantially free of lipid or polymeric nanocarriers. The patent further describes selectable pharmaceutically acceptable carriers and topical formulation formats.
The disclosed topical approach includes potential combination therapy, including combinations with minoxidil and other hair-loss agents. The patent describes evaluation in an in vitro CTLL-2 assay and an in vivo autoimmune alopecia mouse model, with outcomes discussed in relation to hair regrowth using CBD alone versus minoxidil alone versus a CBD+minoxidil combination.
Claims Coverage
The partial set includes two independent claims. Across the claims, the main inventive features are the nanoparticulate cannabidiol formulation with defined purity and particle size, the requirement for skin penetration with substantially free status of lipid or polymeric nanocarriers, and in one claim the addition of minoxidil with a specified ethanol/propylene glycol/water carrier system.
Skin-penetrating nanoparticulate cannabidiol topical composition substantially free of lipid or polymeric nanocarriers
A topical composition consisting of nanoparticulate cannabidiol consisting of greater than 98% by weight cannabidiol and having an average particle size of about 5 nm to about 200 nm, and a pharmaceutically acceptable carrier, wherein the nanoparticulate cannabidiol is capable of penetrating into the skin, wherein the nanoparticulate cannabidiol is substantially free of lipid or polymeric nanocarriers, and wherein the nanoparticulate cannabidiol is present at a concentration of about 5 μM to about 10 μM.
Topical nanoparticulate cannabidiol combined with minoxidil in an ethanol/propylene glycol/water carrier
A topical composition consisting of nanoparticulate cannabidiol consisting of greater than 98% by weight cannabidiol; minoxidil; and a pharmaceutically acceptable carrier consisting of ethanol, propylene glycol, water and optionally one or more of solvent, antisolvent and adjuvant, wherein the cannabidiol is present at a concentration of about 200 g/ml and the minoxidil is present at a concentration of about 5% by weight.
Claim coverage centers on topical compositions that include substantially pure nanoparticulate cannabidiol with a defined average particle size, together with a pharmaceutically acceptable carrier, with skin penetration capability and, in one independent claim, substantially free of lipid or polymeric nanocarriers. A second independent claim adds minoxidil and specifies an ethanol/propylene glycol/water-based carrier with stated concentrations.
Stated Advantages
Enables penetration into the skin.
Documented Applications
Treatment of alopecia areata by administering a topical composition comprising the nanoparticulate cannabidiol of the first independent claim to a subject.
Autoimmune alopecia treatment context is discussed in an in vivo autoimmune alopecia mouse model to evaluate hair regrowth using CBD alone, minoxidil alone, or CBD+minoxidil.
