Abstract
The present disclosure provides redox mediators having two tridentate ligands and analyte sensors comprising such redox mediators. The present disclosure further provides methods of using such analyte sensors for detecting one or more analytes present in a biological sample of a subject.
Core Innovation
The invention relates to an analyte sensor having a distal end with at least a first working electrode and a first active area disposed on a surface of the first working electrode. The first active area is responsive to a first analyte and comprises a first polymer, a first redox mediator covalently bonded to the first polymer, and at least one enzyme responsive to the first analyte. The sensor further includes a mass transport limiting membrane permeable to the first analyte that overcoats at least the first active area.
The first redox mediator is defined by a structural scaffold in which M is iron, ruthenium, osmium, cobalt, or vanadium and n is I, II, III, IV, or V. The substituents R1, R3, R1′, and R3′ are each independently a C1-C12 straight or branched chain alkyl group, and R2 is selected from H and an electron donating group. R2′ is a linking group that covalently bonds the first redox mediator to the first polymer, thereby integrating the redox mediator into the polymer-containing active area.
The invention also includes a method using the analyte sensor by applying a potential to the first working electrode, obtaining a first signal at or above an oxidation-reduction potential of the first active area, and correlating the first signal to the concentration of the first analyte in a fluid contacting the first active area. In further embodiments, additional working electrodes and active areas can be provided for a second analyte, each with its own polymer, covalently bonded redox mediator, and enzyme, and with a corresponding portion of the mass transport limiting membrane overcoating the second active area.
Claims Coverage
The independent claims cover five inventive feature sets centered on an enzyme-responsive active area with a polymer-bound, covalently linked redox mediator and a mass transport limiting membrane, plus a method of electrochemical detection and correlation to analyte concentration. The claims also include a second analyte sensor variant with a second working electrode and second active area responsive to a second, different analyte.
Polymer-bound covalently linked redox mediator active area
An analyte sensor with a first active area responsive to a first analyte, where the first active area comprises a first polymer, a first redox mediator covalently bonded to the first polymer, and at least one enzyme responsive to the first analyte.
Defined redox mediator chemical structure with covalent linking group
The first redox mediator has a structure wherein M is iron, ruthenium, osmium, cobalt, or vanadium; n is I, II, III, IV or V; R1, R3, R1′, and R3′ are each independently a C1-C12 straight or branched chain alkyl group; R2 is selected from H and an electron donating group; and R2′ is a linking group covalently bonding the first redox mediator to the first polymer.
Mass transport limiting membrane overcoating the active area
A mass transport limiting membrane permeable to the first analyte that overcoats at least the first active area.
Electrochemical signal obtained at or above oxidation-reduction potential correlated to concentration
A method including applying a potential to the first working electrode, obtaining a first signal at or above an oxidation-reduction potential of the first active area, the first signal being proportional to a concentration of a first analyte in a fluid contacting the first active area, and correlating the first signal to the concentration of the first analyte in the fluid.
Secondary analyte detection using a second working electrode and second active area
The analyte sensor having a second working electrode with a second active area responsive to a second, different analyte, wherein the second active area includes a second polymer, a second redox mediator covalently bonded to the second polymer, and at least one enzyme responsive to the second analyte, with a second portion of the mass transport limiting membrane overcoating the second active area.
The independent claims are directed to an analyte sensor architecture combining an enzyme-responsive active area with a polymer and a redox mediator covalently bonded to that polymer, a defined redox mediator structure, and a mass transport limiting membrane overcoating the active area. The method claim further covers applying a potential, measuring an electrochemical signal at or above the oxidation-reduction potential, and correlating that signal to analyte concentration.
