Abstract

The present invention relates to novel anti-IgG nanobodies, particularly nanobodies directed against rabbit or mouse IgG. Further, the invention relates to the use of said nanobodies and methods for producing them.

Core Innovation

The invention relates to camelid single-domain antibodies directed against IgG molecules, including single domain antibodies recognizing rabbit or mouse IgG. The reagents are defined by specific sequence identifiers, including CDR3 sequence definitions and full antibody sequences. The single domain antibodies are selected to achieve specificity for a given IgG type and to recognize distinct epitope regions on the IgG molecules.

The invention provides combinations of several different single domain antibodies that recognize the same type of IgG molecules while binding to non-overlapping epitopes on said IgG molecules. Combinations are described that use different single domain antibodies directed to rabbit IgG of SEQ ID NOs. 1 or 2 or to mouse IgG of SEQ ID NOs. 3-34. The combination can comprise a plurality of different single domain antibodies or a plurality of different immune complexes comprising a single domain antibody bound to an IgG antibody.

The disclosed single domain antibodies include specificity by epitope region on IgG molecules, including Fab versus Fc/hinge and light-chain regions. The document describes selection of non-overlapping epitopes across IgG regions such as light chain, kappa and/or lambda light chain, and Fc or hinge fragments. It further provides specific combinations based on defined sequence identifiers for the different single domain antibodies.

Claims Coverage

The independent claims are clm-00001, clm-00010, and clm-00011. Across these independent claims, the coverage is centered on combinations of several single domain antibodies or immune complexes that recognize the same type of IgG but bind non-overlapping epitopes, with further restrictions to specific rabbit or mouse IgG sequence identifiers and, in one case, specific SEQ ID NO pairings.

Non-overlapping epitope IgG-binding multi-nanobody combination

A combination comprising a plurality of different single domain antibodies or a plurality of different immune complexes comprising a single domain antibody bound to an IgG antibody, wherein the single domain antibodies are selected from a combination of several single domain antibodies recognizing the same type of IgG molecules, which bind to non-overlapping epitopes on said IgG molecules.

Rabbit- or mouse-directed IgG binding in a non-overlapping combination

A combination comprising a plurality of different single domain antibodies or a plurality of different immune complexes comprising a single domain antibody bound to an IgG antibody, wherein the single domain antibodies are selected from a combination of several single domain antibodies recognizing the same type of IgG molecules, which bind to non-overlapping epitopes on said IgG molecules, wherein at least one single domain antibody of said combination is directed against rabbit IgG of SEQ ID NOs. 1 or 2; or is directed against mouse IgG of SEQ ID NOs. 3-34.

Specified SEQ ID NO antibody pair combinations with non-overlapping epitope IgG binding

A combination comprising a plurality of different single domain antibodies comprising a single domain antibody bound to an IgG antibody, wherein the single domain antibodies are selected from a combination of several single domain antibodies recognizing the same type of IgG molecules, which bind to non-overlapping epitopes on said IgG molecules, wherein the single domain antibodies are selected from a combination of (i) the single domain antibodies comprising amino acid sequences of SEQ ID NO. 5 and SEQ ID NO. 8; or (ii) the single domain antibodies comprising the amino acid sequence of SEQ ID NO. 8 and SEQ ID NO. 19; or (iii) the single domain antibodies comprising the amino acid sequences of SEQ ID NO. 18 and SEQ ID NO. 19; or (iv) the single domain antibodies comprising the amino acid sequences of SEQ ID NO. 5 and SEQ ID NO. 18; or (v) the single domain antibodies comprising the amino acid sequences of SEQ ID NO. 5 and SEQ ID NO. 25; or (vi) the single domain antibodies comprising the amino acid sequences of SEQ ID NO:1 and SEQ ID NO:2.

The independent claims define combination reagents built from several different single domain antibodies or immune complexes that recognize the same IgG type but bind non-overlapping epitopes, with claim-specific limitations to rabbit or mouse IgG sequence identifiers and, for one independent claim, to specified SEQ ID NO pairings.
