Abstract

The present invention relates to inhibitors of the interaction between H. pylori HopQ and a member of the carcinoembryonic antigen-related cell adhesion molecule (CEACAM) family as well as to immunogenic compositions based on H. pylori HopQ. The present invention further relates to the use of the inhibitors and immunogenic compositions for preventing or treating a disease or disorder caused by or associated with H. pylori.

Core Innovation

Helicobacter pylori (H. pylori) produces an adhesin, HopQ, that interacts with carcinoembryonic antigen-related cell adhesion molecule (CEACAM) family members. The document describes targeting the HopQ–CEACAM interaction to prevent or treat H. pylori-associated diseases and associated gastroduodenal disorders caused by that infection, by interfering with H. pylori-mediated host cell effects.

The document describes inhibitors that block HopQ binding to CEACAMs, including human CEACAM1/3/5/6 and CEACAM N-domain and N-linked fragments. The inhibitors include (poly-)peptide ligands, antibodies and antibody derivatives or mimetics, peptide aptamers, soluble fragments, peptidomimetics, nucleic acids including aptamers and RNA/DNA antisense, and small molecules.

The document also describes immunogenic HopQ-based compositions intended to elicit antibodies that inhibit HopQ–CEACAM binding and downstream signaling. The immunogenic compositions include at least one isolated (poly-)peptide comprising the amino acid sequence of HopQ or an immunogenic fragment comprising an extracellular domain selected from an insertion domain and loops A–D, and immunogenic variants with at least 90% sequence identity to specified sequences, including a HopQ fragment comprising HopQAD with an amino acid sequence corresponding to residues 37 to 463 of SEQ ID NO: 1.

Claims Coverage

The provided content includes an independent claim directed to immunizing against or treating an H. pylori infection by administering a therapeutically effective amount of an immunogenic HopQ-based composition. The inventive features focus on specified HopQ extracellular domains or fragments and their use to raise an immune response or inhibit HopQ–CEACAM interaction.

Immunizing against or treating H. pylori infection with HopQ immunogenic (poly-)peptide or fragment

A method of immunizing against or treating an H. pylori infection by administering a therapeutically effective amount of an immunogenic composition comprising at least one isolated (poly-)peptide comprising the amino acid sequence of H. pylori HopQ, or an immunogenic fragment thereof, where the fragment comprises an extracellular domain selected from an insertion domain and loops A, B, C, and D of HopQ, or an immunogenic variant having at least 90% sequence identity to one of SEQ ID NO: 1, SEQ ID NO: 3, SEQ ID NO: 5 or SEQ ID NO: 15.

Immunizing against or treating H. pylori infection with HopQAD fragment composition

A method where the therapeutically effective amount includes a composition comprising an isolated HopQ fragment comprising HopQAD, wherein HopQAD has an amino acid sequence corresponding to residues 37 to 463 of SEQ ID NO: 1.

Immunizing or treating gastroduodenal disorders caused by H. pylori infection

The method in which an H. pylori infection causes a gastroduodenal disorder, including gastritis, chronic gastritis, gastric or duodenal ulcer, stomach cancer, and MALT lymphoma.

Inhibiting HopQ–CEACAM interaction using an isolated HopQ fragment or immune response

A method where the isolated HopQ fragment or an immune response raised against the fragment inhibits interaction between H. pylori HopQ and a CEACAM family member.

The claim coverage centers on HopQ-based immunogenic compositions comprising specified HopQ extracellular domains or a HopQAD fragment for immunizing against or treating H. pylori infection and associated gastroduodenal disorders, with inhibition of HopQ–CEACAM interaction as a further refinement.
