Abstract
Methods and compositions are provided for treating individuals exhibiting opioid withdrawal symptoms with opioid, benzodiazepine and other drugs of abuse by administering an opioid receptor antagonist agent together with a respiratory stimulant to reverse the effects of opioid withdrawal symptoms, including respiratory depression, sedation, and hypotension. Methods and compositions are also provided for treating stimulant overdose with a benzodiazepine and beta-adrenergic blocking agent. Methods and compositions are also provided for treating concurrent stimulant and opioid overdose comprising administering to a patient in need thereof a benzodiazepine and an opioid antagonist. Methods and compositions are also provided for treating individuals exhibiting opioid withdrawal symptoms or prophy tactically treating individuals for opioid withdrawal symptoms from opioid, benzodiazepine and other drugs of abuse by administering an opioid receptor antagonist agent together with a respiratory stimulant to reverse the effects of withdrawal, including respiratory depression, sedation, and hypotension.
Core Innovation
The invention relates to methods and pharmaceutical compositions for treating opioid overdose in a patient in need thereof by administering an opioid receptor antagonist and a respiratory stimulant. The opioid receptor antagonist is selected from naltrexone, naloxone, nalmefene, and pharmaceutically-acceptable salts thereof, and the respiratory stimulant is selected from a compound of a defined formula or a salt thereof.
The described treatment addresses opioid overdose conditions by reversing respiratory depression, sedation, apnea, and hypotension. The approach additionally covers prophylactically treating opioid withdrawal symptoms by administering the opioid receptor antagonist together with the respiratory stimulant.
The document also describes treating stimulant overdose by administering a benzodiazepine together with a beta-adrenergic blocking agent. For concurrent stimulant/opioid overdose, the document further describes administering a benzodiazepine together with an opioid antagonist.
Administration options include simultaneous or sequential administration, different routes of administration, and separate pharmaceutical compositions. The document additionally includes administration forms such as autoinjectors or emergency kits and depot or controlled-release, including parenteral or nasal formulations.
Claims Coverage
The independent claim covers a method of treating opioid overdose using two coordinated drug components: an opioid receptor antagonist and a respiratory stimulant defined by a structural formula. Dependent claims refine the clinical context and administration parameters, including timing, routes, and whether the antagonist and stimulant are in separate pharmaceutical compositions, and specify an additional named respiratory stimulant compound within the defined formula group.
Opioid antagonist with formula-defined respiratory stimulant for treating opioid overdose
A method of treating opioid overdose by administering to a patient an opioid receptor antagonist selected from naltrexone, naloxone, nalmefene, and pharmaceutically-acceptable salts thereof together with a respiratory stimulant selected from a compound of a following formula or a salt thereof.
Treating opioid overdose where respiratory depression accompanies the overdose
The method includes that the opioid overdose is accompanied by respiratory depression.
Simultaneous or sequential administration of opioid antagonist and respiratory stimulant
The method specifies administering the opioid receptor antagonist and the respiratory stimulant simultaneously or sequentially.
Different routes of administration for antagonist and stimulant
The method specifies that the opioid receptor antagonist and the respiratory stimulant are administered via different routes of administration.
Separate pharmaceutical compositions for antagonist and stimulant
The method specifies administering the opioid receptor antagonist and the respiratory stimulant in separate pharmaceutical compositions.
Named respiratory stimulant compound within the formula group
The method includes using a respiratory stimulant selected from N-(4,6-bis-n-propylamino-[1,3,5]triazin-2-yl)-N,O-dimethyl-hydroxylamine, and a pharmaceutically-acceptable salt thereof.
Overall claim coverage centers on treating opioid overdose by combining a specified opioid receptor antagonist with a respiratory stimulant defined by a structural formula, with dependent claims narrowing to respiratory depression, timing, delivery routes, separate pharmaceutical compositions, and a named respiratory stimulant compound.
