Abstract
The present disclosure provides herein anti-CD4 antibodies or antigen-binding fragments thereof, isolated polynucleotides encoding the same, pharmaceutical compositions comprising the same, and the uses thereof.
Core Innovation
The disclosed method eliminates CD4+ T cells in a subject in need thereof by administering an effective amount of an antibody, or an antigen-binding fragment thereof, that specifically binds to human CD4. The antibody or fragment contains one of five specified combinations of heavy- and light-chain variable-region CDRs, including antibodies designated 2B6, 29B7, 33E12, 11E12, and 10C2.
Humanized and chimeric anti-CD4 antibodies bind human CD4 and induce antibody-dependent cellular cytotoxicity, producing approximately 85–90% CD4+ cell depletion in peripheral blood mononuclear cells along with NK-cell-mediated ADCC. Because the background section is omitted from the disclosure, the problem being solved is not explicitly described in patent.
Claims Coverage
The claim information identifies one independent claim directed to eliminating CD4+ T cells in a subject using an antibody or antigen-binding fragment specifically binding human CD4, comprising one inventive feature.
Elimination of CD4+ T cells using specified anti-CD4 antibodies
The method comprises administering to a subject in need thereof an effective amount of an antibody specifically binding to human CD4, or an antigen-binding fragment thereof, containing one of five specified combinations of heavy- and light-chain CDRs.
The independent claim provides a method of eliminating CD4+ T cells via administration of a human CD4-binding antibody or antigen-binding fragment having one of the specified CDR combinations, with dependent claims further reciting antibody formats, conjugates, functional properties, and cancer-related conditions.
Stated Advantages
Mutations in the human IgG1 constant region are described as increasing CDC or ADCC relative to the wild-type human IgG1 constant region.
The described antibodies and variants exhibit CD4 binding and ADCC activity.
Documented Applications
Eliminating CD4+ T cells in a subject in need thereof using an antibody or antigen-binding fragment that specifically binds human CD4.
Treating subjects with cancer or cancer-related conditions, including solid tumors, leukemia, myeloma, lymphoma, myelodysplastic syndrome, sarcoma, and teratoma.
