Abstract

Provided is a method for regenerating a liquid to be treated, the method includes bringing a lactic acid adsorbent that contains a Mg—Al-based layered double hydroxide having Mg2+ and Al3+ as constituent metals, and an ammonia adsorbent having L-type zeolite, into contact with the liquid to be treated that contains lactic acid and ammonia, to remove lactic acid and ammonia in the liquid to be treated.

Core Innovation

The regeneration method regenerates a liquid to be treated that contains lactic acid and ammonia by bringing a lactic acid adsorbent and an ammonia adsorbent into contact with the liquid to be treated. The lactic acid adsorbent contains a Mg–Al-based layered double hydroxide having Mg2+ and Al3+ as constituent metals, and the ammonia adsorbent has L-type zeolite. The approach removes lactic acid and ammonia from the culture liquid.

The liquid to be treated is a culture liquid of at least either cell or microorganism that contains glucose. The method adjusts an amount of consumption of each of the lactic acid adsorbent and the ammonia adsorbent such that a concentration thereof relative to the liquid to be treated is 0.05 g/mL or higher and 0.1 g/mL or lower for both the lactic acid adsorbent and the ammonia adsorbent. The described regeneration suppresses pH fluctuation and is evaluated for selectivity with respect to glucose.

The regeneration is described as being implemented as simultaneous contact treatment and also as sequential treatment in a two-step manner, and the adsorbents are evaluated by screening of various adsorbent combinations. The document concludes that only the combination of Mg–Al-based layered double hydroxide and L-type zeolite achieves consistently high lactic acid and ammonia adsorption across the simultaneous and sequential processes.

Claims Coverage

The document provides two independent claims that define complementary coverage: a regeneration method and a regenerating agent. Across both independent claims, there are three main inventive feature clusters: the paired adsorbent materials (Mg–Al-based layered double hydroxide for lactic acid and L-type zeolite for ammonia), the nature of the liquid to be treated as a glucose-containing culture liquid, and the adjusted consumption amounts specified as concentration relative to the liquid within 0.05 g/mL to 0.1 g/mL for both adsorbents.

Paired adsorbents for lactic acid and ammonia removal

A lactic acid adsorbent that contains a Mg–Al-based layered double hydroxide having Mg2+ and Al3+ as constituent metals, and an ammonia adsorbent having L-type zeolite, brought into contact with the liquid to be treated that contains lactic acid and ammonia to remove lactic acid and ammonia.

Glucose-containing culture liquid as the liquid to be treated

The liquid to be treated is a culture liquid of at least either cell or microorganism that contains glucose.

Adjusted adsorbent consumption concentration range

An amount of consumption of each of the lactic acid adsorbent and the ammonia adsorbent is adjusted so that a concentration thereof relative to the liquid to be treated is 0.05 g/mL or higher and 0.1 g/mL or lower for both the lactic acid adsorbent and the ammonia adsorbent.

Claim coverage centers on contacting (in the method) or providing (in the regenerating agent) a specific adsorbent pair—Mg–Al-based layered double hydroxide and L-type zeolite—to remove both lactic acid and ammonia from a glucose-containing culture liquid, with the consumption amounts constrained so that both adsorbents have a concentration relative to the liquid between 0.05 g/mL and 0.1 g/mL.

Stated Advantages

Lower volume/cost and simplified device structure compared with dialysis.

High adsorption performance for lactic acid with selectivity over glucose.

High adsorption performance for ammonia with selectivity over glucose.

Suppression of pH fluctuation (controlled pH change).

Documented Applications

Regeneration of a culture liquid containing lactic acid and ammonia for at least either cells or microorganisms, where the culture liquid contains glucose.

Lactic-acid-first and ammonia-first sequential two-step treatment modes and simultaneous contact treatment modes for removing lactic acid and ammonia from culture liquids.

Use of adsorbent screening to identify that the Mg–Al-based layered double hydroxide plus L-type zeolite combination achieves consistently high adsorption across simultaneous and sequential processes.
