Abstract

The present invention relates to an antibacterial protein CDL200 with an antibacterial activity specific against Clostridioides difficile. More specifically, the present invention relates to an antibacterial protein CDL200 specific to Clostridioides difficile that may infect and cause disease in animals including humans, the antibacterial protein CDL200 being characterized by having the ability to specifically lyse the Clostridioides difficile and containing the amino acid sequence represented by SEQ ID NO: 1. In addition, the present invention relates to a pharmaceutical composition for treating an infection or disease caused by Clostridioides difficile, the composition containing the antibacterial protein CDL200 specific to Clostridioides difficile as an active ingredient.

Core Innovation

The invention relates to an antibacterial protein, CDL200, that has Clostridioides difficile-specific antibacterial activity. The protein comprises the amino acid sequence represented by SEQ ID NO: 1 and is characterized as specifically lytic against Clostridioides difficile.

The invention addresses prevention and treatment of Clostridioides difficile infection or disease through a pharmaceutical composition that includes CDL200 as an active ingredient. The composition is described as obtaining selective lysis of C. difficile and reduced impact on normal intestinal flora.

The invention further addresses antibacterial activity relevant to antibiotic-resistant C. difficile. Documented evaluations in the provided content reference in vitro lysis-relevant assays, including a liquid lysis test and a turbidity reduction assay, and an animal infection model described with survival rate and corresponding figures.

Claims Coverage

The document provides three independent claims directed to a Clostridioides difficile-specific antibacterial protein defined by SEQ ID NO: 1, treatment of a subject with a C. difficile infection or disease by administering a composition containing that protein, and killing an infected cell by contacting the cell with that protein composition. Across the independent claims, two inventive feature themes are present: a defined protein sequence and use of a composition containing that protein for treatment or killing.

Clostridioides difficile-specific antibacterial protein CDL200 comprising SEQ ID NO: 1

A Clostridioides difficile-specific antibacterial protein CDL200 having specific antibacterial activity to Clostridioides difficile and comprising the amino acid sequence represented by SEQ ID NO: 1.

Treating a subject by administering a CDL200 composition

Administering to a subject having a Clostridioides difficile infection or disease a composition comprising a Clostridioides difficile-specific antibacterial protein CDL200 having specific antibacterial activity to Clostridioides difficile and comprising the amino acid sequence represented by SEQ ID NO: 1.

Killing infected cells by contacting with a CDL200 composition

Killing a cell infected with Clostridioides difficile by contacting the cell with a composition comprising a Clostridioides difficile-specific antibacterial protein CDL200 having specific antibacterial activity to Clostridioides difficile and comprising the amino acid sequence represented by SEQ ID NO: 1.

Overall, the independent claims focus on CDL200 defined by the amino acid sequence of SEQ ID NO: 1, and on using a composition containing that defined protein for treating C. difficile infection or disease or killing cells infected with C. difficile.

Stated Advantages

Selective lysis of C. difficile with minimal effect on normal intestinal flora.

Activity against antibiotic-resistant C. difficile.

Documented Applications

Preventing or treating a Clostridioides difficile infection or disease in a subject, including humans.

Use of CDL200 in an animal infection model, with reported survival rate and referenced lysis-relevant assay outcomes, including a liquid lysis test and a turbidity reduction assay.
