Abstract

The invention provides compositions and methods for inhibiting the interaction of the SARS-CoV-2 S protein and one or more SARS-CoV-2 S protein interacting protein, and methods of use thereof.

Core Innovation

The invention relates to SARS-CoV-2 Spike (S) protein engaging SARS-CoV-2 S protein interacting partner(s) beyond ACE2, including immune-cell receptors/interaction partners comprising C-type lectins (DC-SIGN, L-SIGN, LSECtin, ASGR1, CLEC10A) and TTYH2. The described binding is largely mediated via non-RBD epitopes, and this interaction is associated with hyperinflammatory COVID-19.

The invention provides inhibitors that block or reduce binding of SARS-CoV-2 S protein to one or more SARS-CoV-2 S protein interacting partner(s). The inhibitor embodiments include antibodies/nanobodies, decoy receptors, Fc fusions, and bispecific/multispecific antibodies, as well as nucleic acids and other agents, with optional detecting of S protein.

Overall, the invention centers on inhibiting SARS-CoV-2 S protein binding to selected S protein interacting partner(s), where the interacting partners include C-type lectins and TTYH2. In particular, certain embodiments provide a bispecific antibody inhibitor defined by specific CDR sequences corresponding to SEQ ID NO: 57–SEQ ID NO: 59 and SEQ ID NO: 93–SEQ ID NO: 95. This binding-blocking strategy is presented as a way to treat and/or prevent SARS-CoV-2 infection and related diseases, with optional detection of S protein.

Claims Coverage

The partial claim set includes one independent claim. The independent claim covers an inhibitor defined as a bispecific antibody with CDR sequences from specific SEQ ID NO ranges, targeting inhibition of SARS-CoV-2 S protein binding to one or more SARS-CoV-2 S protein interacting partners.

Bispecific antibody inhibitor of S protein interacting-partner binding defined by specific CDR sequences

An inhibitor of the binding of SARS-CoV-2 S protein to one or more SARS-CoV-2 S protein interacting partner(s), wherein the inhibitor is a bispecific antibody comprising the CDR sequences of SEQ ID NO: 57–SEQ ID NO: 59 and the CDR sequences of SEQ ID NO: 93–SEQ ID NO: 95.

The claim coverage, as provided, is anchored by the independent claim requiring an inhibitor that blocks SARS-CoV-2 S protein binding to one or more interacting partners, specifically through a bispecific antibody defined by CDR sequence ranges SEQ ID NO: 57–SEQ ID NO: 59 and SEQ ID NO: 93–SEQ ID NO: 95.

Stated Advantages

Documented Applications

Treating COVID-19 by administering the inhibitor or a nucleic acid molecule encoding the inhibitor.

Detecting SARS-CoV-2 S protein in a sample by contacting the sample with the inhibitor (or a composition comprising it) and detecting the presence of SARS-CoV-2 S protein.
