Abstract

The present invention relates to methods for detecting analytes in a sample. The methods may also be used to determine the amount of analyte in the sample. The present invention also relates to antibody or antibody-like molecules and labelled antigens for use in these methods.

Core Innovation

The invention provides a method for detecting an analyte in a sample using chemiluminescence resonance energy transfer (CRET). The sample is contacted with a first molecule comprising an antibody or antibody-like molecule capable of binding to the analyte attached to a first component of a CRET pair, and a second molecule comprising a labelled antigen attached to a second component of the CRET pair. Energy transfer occurs between the first component and the second component when the labelled antigen is bound to the antibody or antibody-like molecule.

The CRET pair is configured such that the first component comprises a luciferase and the second component comprises a fluorescent protein, or the first component comprises a fluorescent protein and the second component comprises a luciferase. When the labelled antigen is bound, the efficiency of energy transfer is in the range of 10 to 75%, and detecting the analyte is performed by detecting that the efficiency of energy transfer has been altered in the presence of the sample, where a reduction in the efficiency indicates that the analyte is present.

The disclosure further provides a CRET pair solution comprising an antibody or antibody-like molecule and a labelled antigen, where the CRET pair comprises luciferase and fluorescent protein in either order and the energy transfer efficiency is 10 to 75% when the labelled antigen is bound. The solution includes an antibody or antibody-like molecule attached to a first component of the CRET pair and capable of binding to the analyte, together with a labelled antigen comprising an antigen capable of binding to the antibody or antibody-like molecule attached to a second component of the CRET pair.

Claims Coverage

The independent claims cover a method for detecting an analyte and CRET solution compositions. The claims are centered on a CRET-based detection scheme using an antibody or antibody-like molecule and a labelled antigen, with luciferase and fluorescent protein configured in either order and an energy transfer efficiency of 10 to 75% when the labelled antigen is bound.

Antibody or antibody-like molecule CRET detection with labelled antigen

A method for detecting an analyte in a sample by contacting the sample with a first molecule comprising an antibody or antibody-like molecule attached to a first component of a CRET pair and a second molecule comprising a labelled antigen attached to a second component of the CRET pair, where energy transfer between the first and second components occurs when the labelled antigen is bound to the antibody or antibody-like molecule.

Luciferase and fluorescent protein CRET pair arrangement

The CRET pair is arranged so that the first component comprises a luciferase and the second component comprises a fluorescent protein, or the first component comprises a fluorescent protein and the second component comprises a luciferase.

10 to 75% energy transfer efficiency and reduction indicates analyte presence

When the labelled antigen is bound, the efficiency of energy transfer between the first component and the second component is in the range of 10 to 75%, and detecting an altered efficiency of energy transfer in the presence of the sample includes that a reduction in the efficiency indicates the analyte is present.

CRET pair solution with antibody or antibody-like molecule and labelled antigen

A solution comprising an antibody or antibody-like molecule capable of binding to an analyte attached to a first component of a CRET pair and a labelled antigen comprising an antigen capable of binding to the antibody or antibody-like molecule attached to a second component of the CRET pair, where the CRET pair comprises luciferase and fluorescent protein in either order.

10 to 75% energy transfer efficiency in CRET solution

When the labelled antigen is bound, the efficiency of energy transfer between the first component and the second component is in the range of 10 to 75%.

Across the independent claims, the inventive coverage is consistently focused on a CRET pair-based system using an antibody or antibody-like molecule and a labelled antigen, with luciferase and fluorescent protein configured in either order and a defined energy transfer efficiency range of 10 to 75% when the labelled antigen is bound.
