Abstract

Methods for treating cancer and/or inducing tumor regression in mammals (e.g., humans) by increasing the metabolism of the mammal, administering a BA dye to the mammal, and thereafter exposing the tumor to actinic light for activation of the BA dye.

Core Innovation

The invention relates to a method for treating a pancreatic cancer tumor in a subject by intentionally raising plasma glucose above a basal plasma glucose level and then performing photosensitizer-based photodynamic tumor treatment. The method administers at least one of glucose, glucagon, corticosteroids, growth hormones, thyroid hormones and insulin to raise the plasma glucose level above the basal level. Thereafter, at least one photosensitizer selected from benzophenoxazinium, benzophenothiazinium, and benzophenoselenazinium dyes is administered to the subject.

After the photosensitizer is administered, the tumor is exposed to actinic light after the plasma glucose level has been raised to a level above the basal plasma glucose level. In one formulation, the exposing occurs between about 10 minutes and one week after raising the plasma glucose level above basal. In another formulation, the plasma glucose level is maintained above the basal level until exposure of the tumor to actinic light, and the exposing occurs after the plasma glucose level has been raised above basal.

The treatment is further defined by selecting the photosensitizer dyes within the benzophenoxazinium/benzophenothiazinium/benzophenoselenazinium group and by specifying exposure timing and actinic light parameters. The actinic light is characterized as having a wavelength range of about 600 nm to about 700 nm in at least one formulation. Additional constraints specify total light energy in a range of about 50 to about 500 Joules per square centimeter.

Claims Coverage

The partial record contains two independent claims directed to treating pancreatic cancer by combining glucose elevation with benzophenoxazinium/benzophenothiazinium/benzophenoselenazinium photosensitizers and subsequent exposure to actinic light, with at least one claim adding timing and actinic-light constraints.

Raising plasma glucose above basal before actinic exposure

administering at least one of glucose, glucagon, corticosteroids, growth hormones, thyroid hormones and insulin, to the subject until the plasma glucose level of the subject is raised to a level above the basal plasma glucose level of the subject; maintaining the plasma glucose level of the subject above the basal plasma glucose level of the subject until exposure of the tumor to actinic light

Administering benzophenoxazinium/benzophenothiazinium/benzophenoselenazinium photosensitizer

administering at least one photosensitizer selected from the group consisting of benzophenoxazinium, benzophenothiazinium, and benzophenoselenazinium dyes to the subject

Exposing the tumor to actinic light after glucose elevation

exposing the tumor to actinic light between about 10 minutes and one week after the plasma glucose level of the subject has been raised to a level above the basal plasma glucose of the subject

Actinic light in defined spectral range

using actinic light that has a wavelength in the range of 600–700 nm

Actinic light with defined total light energy

providing total light energy in the range of about 50 to about 500 Joules per square centimeter

Across the independent claims, the main inventive combination is raising plasma glucose above basal using selected glucose-related agents, administering benzophenoxazinium/benzophenothiazinium/benzophenoselenazinium photosensitizer dyes, and exposing the pancreatic cancer tumor to actinic light after glucose is raised, with maintenance of elevated glucose in one claim, and with timing and actinic-light spectral and energy parameters in one claim.
