Abstract

The present invention relates to peptides, proteins, nucleic acids and cells for use in immunotherapeutic methods. In particular, the present invention relates to the immunotherapy of cancer. The present invention furthermore relates to tumor-associated T-cell peptide epitopes, alone or in combination with other tumor-associated peptides that can for example serve as active pharmaceutical ingredients of vaccine compositions that stimulate anti-tumor immune responses, or to stimulate T cells ex vivo and transfer into patients. Peptides bound to molecules of the major histocompatibility complex (MHC), or peptides as such, can also be targets of antibodies, soluble T-cell receptors, and other binding molecules.

Core Innovation

The disclosure provides tumor-associated peptide epitopes derived from tumor-associated genes and naturally presented by HLA on primary lung cancer tissues, including NSCLC and SCLC. It also identifies and selects MHC-restricted tumor-associated peptides and defines peptide targets by MHC molecule together with peptide, with recognition by CD8 T cells.

The document describes peptides in the form of pharmaceutically acceptable salts, including RWMDQRLVF (SEQ ID NO: 55), and provides defined peptide sequences and associated HLA allotypes such as HLA-A*01, HLA-A*02, HLA-A*03, HLA-A*24, HLA-B*07, HLA-B*08, and HLA-B*44. It further addresses peptide variants, elongated peptides, and class II peptides, together with allele population coverage.

The disclosure also describes cell-SELEX selection of aptamers that bind to MHC/peptide complexes, antibodies against MHC/peptide complexes including recombinant polyclonal, monoclonal, bispecific, and chimeric antibodies, and recombinant antibody production involving immunizing a genetically engineered non-human mammal expressing human MHC I/II, followed by mRNA isolation and phage display library screening. The peptides are proposed for immunotherapy use, including vaccine and cell-based therapies, medicament applications, and radionuclide imaging such as PET imaging and therapy centered on antigen-loading and killing of tumor cells.

Claims Coverage

The provided independent claims cover the specific peptide sequence RWMDQRLVF (SEQ ID NO: 55) in pharmaceutically acceptable salt form, and a pegylated embodiment of the same sequence. Across the independent-claim level, the inventive focus is on the defined peptide identity together with salt and pegylation embodiments for immunotherapy-related use.

Pharmaceutically acceptable salt peptide of SEQ ID NO:55

A peptide consisting of the amino acid sequence RWMDQRLVF (SEQ ID NO:55) in the form of a pharmaceutically acceptable salt.

Pegylated peptide of SEQ ID NO:55

A pegylated peptide consisting of the amino acid sequence of RWMDQRLVF (SEQ ID NO:55) or a pharmaceutically acceptable salt thereof.

MHC class-I binding with CD8 T cell recognition

The peptide is configured to bind an MHC class-I molecule and, when bound, is recognized by CD8 T cells.

Chloride salt form

The peptide is present as a pharmaceutically acceptable chloride salt.

Pharmaceutical composition with water and buffer

A pharmaceutical composition includes the peptide and is formulated with water and a buffer.

Adjuvant selected from an enumerated immunostimulatory set

The composition further includes an adjuvant selected from an enumerated list of immunostimulatory agents and related formulation components.

Peptide produced by solid phase synthesis or yeast/bacterial expression

The peptide, present as a pharmaceutically acceptable salt, is produced either by solid phase peptide synthesis or via expression in a yeast cell or bacterial cell.

Claim coverage is centered on the defined peptide sequence RWMDQRLVF (SEQ ID NO: 55), with independent-claim scope established by pharmaceutically acceptable salt and pegylated embodiments. Dependent features add MHC class-I binding and CD8 T cell recognition, chloride salt form, formulation elements, optional adjuvants, and production routes.

Stated Advantages

Not explicitly described in patent.
