Abstract

The present invention relates to transgenic ornamental barbs, as well as methods of making such fish by in vitro fertilization techniques. Also disclosed are methods of establishing a population of such transgenic barbs and methods of providing them to the ornamental fish industry for the purpose of marketing.

Core Innovation

The invention relates to transgenic Barb comprising chromosomally integrated expression cassettes that encode fluorescent proteins for ornamental and research use. In particular, the Barb comprises an “Orange Barb 1 transformation event” in which sperm comprising the Orange Barb 1 transformation event has been deposited at the ATCC as deposit no. PTA-126571, and a “Purple Barb 1 transformation event” deposited at the ATCC as deposit no. PTA-126570. The integrated expression cassette(s) are inherited such that sperm, eggs, embryos, and progeny inherit the integrated events.

The invention further defines the fertility and zygosity behavior of the transgenic Barbs carrying the integrated events, including fertile transgenic Barbs and variant genetic states such as homozygous, heterozygous, and hemizygous. Progeny derived from breeding are described as carrying the chromosomally integrated transformation event, with fertility and inheritance used to establish populations of the transgenic Barbs.

The invention also includes production and distribution of additional transgenic Barbs by breeding with a second Barb, where the second Barb can be transgenic or non-transgenic. For market placement, the transgenic Barbs are distributed into the ornamental fish market through growers, commercial distributors, and multi-product retailers with ornamental fish departments, including fluorescence-based identification during visual inspection.

Stated uses include ornamental fish applications and scientific research applications enabled by fluorescent transgenic fish, including cell lineage tracing, cell migration, genetic mosaic marking, and fish cancer models.

Claims Coverage

The document includes two independent claims covering a transgenic Barb containing a specific chromosomally integrated fluorescent protein expression cassette associated with an ATCC-deposited transformation event, and a method of producing transgenic Barbs by obtaining such a Barb and breeding it with a second Barb. Across these independent claims, the inventive features center on the Orange Barb 1 transformation event, chromosomal integration of an orange fluorescent protein expression cassette, and breeding with a second Barb to generate additional Barb carrying the same transformation event.

Chromosomally integrated orange fluorescent protein expression cassette with Orange Barb 1 event

A transgenic Barb comprising a chromosomally integrated expression cassette encoding an orange fluorescent protein, wherein the Barb comprises an “Orange Barb 1 transformation event,” with sperm comprising the Orange Barb 1 transformation event having been deposited at the ATCC as deposit no. PTA-126571.

Producing a transgenic Barb by obtaining Orange Barb 1 event carrier and breeding with a second Barb

A method of producing a transgenic Barb comprising obtaining a Barb that comprises a chromosomally integrated expression cassette encoding an orange fluorescent protein, wherein the Barb comprises an “Orange Barb 1 transformation event,” sperm comprising the Orange Barb 1 transformation event having been deposited at the ATCC as deposit no. PTA-126571; and breeding the obtained Barb with a second Barb to provide a transgenic Barb comprising the Orange Barb 1 transformation event.

Overall claim coverage is directed to transgenic Barbs that carry a chromosomally integrated expression cassette for an orange fluorescent protein associated with the deposited “Orange Barb 1 transformation event,” and to a production method that obtains such a carrier and breeds it with a second Barb to provide additional transgenic Barbs comprising the Orange Barb 1 transformation event.
