Abstract

The embodiments disclosed herein generally relate to systems, devices and methods for the fractionation, isolation, extraction and processing of the adipose supernatant layer of a bone marrow aspirate. In particular, the various embodiments relate to systems, devices, and methods of obtaining, utilizing, and processing the adipose supernatant layer of a bone marrow aspirate as a source of mesenchymal stem cells.

Core Innovation

The disclosed subject matter provides a fluid processing device with first and second adjacent chambers that share a shared sidewall portion. An opening extends through the shared sidewall from a vertical inner surface of the first chamber to a vertical inner surface of the second chamber, defining a fluid pathway between the chambers.

The device includes a mechanical emulsification system in fluid communication with the second chamber for emulsifying a liquid contained in the second chamber. The document also describes structural and functional refinements such as locating the opening at an upper edge, adding syringe-connectable connectors to one or both upper openings, and fixing an emulsification grate to the second sidewall.

A related method processes a fluid using the device by placing the fluid into the first chamber, centrifuging to separate fluid into fluid fractions, and decanting one or more fluid fractions from the first chamber into the second chamber through the opening while leaving one or more fluid fractions in the first chamber. The method includes re-centrifuging the fluid fractions in the first and second chambers and removing one or more fluid fractions from at least one of the first chamber and the second chamber, with mechanical emulsification applied to a liquid in the second chamber.

In the broader described application context, the fluid includes density-fractionated bone marrow aspirate and emphasizes collecting and processing the adipose supernatant layer, adipose layer supernatant or adipose LS, rather than discarding it. The document describes optional co-processing of buffy coat and emphasizes mechanical emulsification to disrupt the adipose collagen/lipid matrix to increase mesenchymal stem cells, with later therapeutic closed-loop reintroduction and optional culture expansion described in the disclosed examples.

Claims Coverage

The document includes two independent claims: one directed to a fluid processing device and one directed to a method for processing a fluid. Each independent claim centers on a two-chamber structure with a shared sidewall and an opening defining a fluid pathway, combined with a mechanical emulsification system in the second chamber; the method claim additionally recites fraction separation and selected transfer/removal steps after centrifugation.

Shared sidewall with inter-chamber opening

The device includes first and second adjacent chambers with a shared sidewall portion, wherein a first surface of the shared sidewall is a vertical inner surface of the first chamber and a second surface of the shared sidewall is a vertical inner surface of the second chamber, and wherein the opening extends through the shared sidewall from the first surface to the second surface, defining a fluid pathway from the first chamber to the second chamber.

Mechanical emulsification system in the second chamber

The device includes a mechanical emulsification system in fluid communication with the second chamber for emulsifying a liquid contained in the second chamber.

Two-chamber processing with centrifugation, decanting, re-centrifugation, and removal

The method provides the device with first and second adjacent chambers sharing a sidewall portion and an opening extending through the shared sidewall to define a fluid pathway, places the fluid into the first chamber, centrifuges to separate the fluid into fluid fractions, decants one or more fluid fractions from the first chamber into the second chamber through the opening while leaving one or more fluid fractions in the first chamber, re-centrifuges the fluid fractions in the first and second chambers, and removes one or more fluid fractions from at least one of the first chamber and the second chamber.

Mechanical emulsification applied during second-chamber processing

The method recites that the device includes a mechanical emulsification system in fluid communication with the second chamber for emulsifying a liquid contained in the second chamber, with the above centrifugation, decanting, re-centrifugation, and removal steps performed using the device.

Overall claim coverage centers on a two-chamber device architecture using a shared sidewall with an opening that defines a fluid pathway to transfer selected fractions, and a mechanical emulsification system configured to emulsify a liquid in the second chamber. The method claim further specifies centrifuging to separate fractions, decanting selected fractions into the second chamber through the shared-sidewall opening, re-centrifuging, and removing fractions from at least one chamber.
