Abstract
Described herein are particles comprising a first compartment, a second compartment, and a compound of Formula (I), as well as compositions and methods of making and using the same. The particles may comprise a cell capable of expressing a therapeutic agent useful for the treatment of a disease, disorder, or condition described herein.
Core Innovation
The invention provides particles comprising a first compartment and a second compartment. The first compartment comprises an alginate polymer and a cell, including islet cells, and the second compartment comprises an alginate polymer and surrounds the first compartment. The alginate polymer of the second compartment is modified with a compound of Formula (I-a), Formula (III-a), or Formula (I), or a pharmaceutically acceptable salt thereof.
The disclosure defines broad structural variability for the modifying compound and describes chemically modified alginate in which the Formula compound is chemically conjugated to alginate, with optional disposition on an exterior surface. The embodiments also set out sub-formula embodiments and example compounds, including Compounds 100-104 and other listed compounds.
The particles are described as encapsulating engineered cells that express therapeutic agents, including replacement proteins such as Factor VIII and Factor IX variants. The document also describes cell-type scope including epithelial cells, endothelial cells, fibroblast cells, mesenchymal stem cells, keratinocytes, islet cells, and stem/iPS-derived cells.
Claims Coverage
The consolidated claim set includes two independent claims. They cover a particle with a cell-containing first compartment surrounded by a second alginate compartment, and the second compartment’s alginate is modified with a formula-defined compound.
Two-compartment alginate particle with cell-containing first compartment
A particle comprising a first compartment comprising an alginate polymer and a cell, including an islet cell, and a second compartment comprising an alginate polymer, wherein the first compartment is surrounded by the second compartment.
Alginate modified with formula-defined compound
The alginate polymer of the second compartment is modified with a compound of Formula (I-a), Formula (III-a), or Formula (I), or a pharmaceutically acceptable salt thereof, with broad structural definitions for the modifying compound.
Overall, the independent claims define a two-compartment alginate particle architecture and tie that architecture to chemical modification of the second-compartment alginate with a formula-defined compound.
Stated Advantages
More afibrotic behavior characterized by reduced foreign body response (FBR) and reduced fibrosis.
Maintaining capsule integrity.
Documented Applications
Encapsulation of engineered cells producing replacement proteins such as Factor VIII and Factor IX variants for therapeutic delivery.
