Abstract
Presented herein, in certain aspects, are cell-free therapeutic composition derived from human amniotic fluid (HAF) and uses thereof for the prevention and treatment of selected diseases and disorders.
Core Innovation
The invention relates to cell-free therapeutic compositions derived from modified human amniotic fluid (HAF) and comprising HAF-derived nanoparticles. The HAF-derived nanoparticles have a diameter of between 50 and 200 nm and are present at a concentration of at least 1×10^11 nanoparticles/ml in the composition.
The modified HAF is used as the starting material to provide the nanoparticles for therapeutic administration to a subject. The compositions are characterized using defined physicochemical properties, including exclusion of cells/pathogens and removal of unwanted material, and may include optional hyaluronic acid.
The nanoparticles or exosomes are also described with defined nanoparticle cargo, including specified surface proteins and microRNAs selected from Table 1. The disclosed therapeutic approaches include methods of treating disease or disorder in a subject by administering a therapeutically effective amount of the modified HAF and HAF-derived nanoparticles composition.
Respiratory indications described include acute respiratory syndrome/ARDS, COPD, and bronchopulmonary dysplasia, and other described indications include osteoarthritis/rheumatoid arthritis and acute brain injury and acute organ failure associated with COVID-19.
Claims Coverage
The relevant independent claims cover methods of treating a disease or disorder and a respiratory disorder, each defined by administration of a therapeutically effective amount of a composition comprising modified HAF and HAF-derived nanoparticles with specific diameter and concentration constraints. Two inventive features are central across the independent claims.
Therapeutic composition with modified HAF and defined nanoparticle size
Administering to the subject a therapeutically effective amount of a composition comprising a modified human amniotic fluid (HAF) and HAF-derived nanoparticles, wherein the nanoparticles have a diameter of between 50 and 200 nm.
Therapeutic composition with modified HAF and defined nanoparticle concentration
Administering to the subject a therapeutically effective amount of a composition comprising a modified human amniotic fluid (HAF) and HAF-derived nanoparticles, wherein the concentration of the nanoparticles in the composition is at least 1×10^11 nanoparticles/ml.
Treatment of a disease or disorder using modified HAF nanoparticle composition
A method of treating a disease or disorder in a subject comprising administering to the subject a therapeutically effective amount of a composition comprising modified human amniotic fluid (HAF) and HAF-derived nanoparticles with a diameter of between 50 and 200 nm and a concentration of the nanoparticles in the composition being at least 1×10^11 nanoparticles/ml.
Treatment of a respiratory disorder using modified HAF nanoparticle composition
A method of treating a respiratory disorder in a subject comprising administering to the subject a therapeutically effective amount of a composition comprising modified human amniotic fluid (HAF) and HAF-derived nanoparticles with a diameter of between 50 and 200 nm and a concentration of the nanoparticles in the composition being at least 1×10^11 nanoparticles/ml.
Overall, the independent claim coverage is anchored in administering a therapeutically effective amount of a composition comprising modified HAF and HAF-derived nanoparticles with a 50–200 nm diameter range and at least 1×10^11 nanoparticles/ml concentration, with one independent claim broadly covering any disease or disorder and the other focusing on respiratory disorders.
