Abstract

Described herein is an adaptive expert system comprising a computing device having a memory that stores programmatic instructions and a processor that executes the programmatic instructions. The adaptive expert system receives sample data comprising at least one of raw data, optical data, and electropherogram data from a DNA analysis device, said data generated from a sample containing DNA. The adaptive expert system determines at least one characteristic of the sample data. The adaptive expert system utilizes the at least one characteristic to classify the sample data and apply a predefined parameter set to the said sample data to generate an output.

Core Innovation

A system receives sample data comprising raw data, optical data, and electropherogram data from a DNA analysis device that generates the sample data from a sample containing DNA. The system determines at least one characteristic of the sample data, including signal strength, and utilizes the at least one characteristic to classify the sample data. The system applies a predefined parameter set to the sample data to generate a plurality of structured data files comprising a DNA ID, with the number of structured data files generated based upon the signal strength.

A computing device performs, for each of the plurality of structured data files, a series of searches for DNA data matching one or more characteristics of the DNA ID. The matching uses progressively less conservative match criteria in each search, and each search generates a match score, compares the match score to a predetermined threshold score, and transmits the match score and the matched DNA data for display when the match score meets or exceeds the predetermined threshold score.

When none of the match scores for the series of searches meet or exceed the predetermined threshold score, the system automatically iterates one or more values in the predefined parameter set based upon the match scores to generate a second predefined parameter set. The system then applies the second predefined parameter set to generate a plurality of second structured data files comprising a second DNA ID and performs a second series of searches using progressively less conservative match criteria, repeating the iteration-and-search sequence across DNA IDs to produce search matches for display.

Claims Coverage

The independent claim coverage includes three inventive features, with additional refinements describing mixture-triggered parameter modifications, visualization output generation, and match-score definition via specified metrics.

Signal-strength-based classification and multiple structured outputs

determine at least one characteristic including signal strength to classify the sample data and apply a predefined parameter set to generate a plurality of structured data files comprising a DNA ID, wherein the number of structured data files generated is based upon the signal strength

Progressively less conservative iterative DNA searches with thresholding and display

perform a series of searches for DNA data matching one or more characteristics of the DNA ID using progressively less conservative match criteria, generating a match score, comparing the match score to a predetermined threshold score, and transmitting the match score and matched DNA data for display when the match score meets or exceeds the predetermined threshold score

Automatic parameter-set iteration triggered by search-match failure

when none of the match scores meet or exceed the predetermined threshold score, automatically iterate one or more values in the predefined parameter set based upon the match scores to generate a second predefined parameter set, apply the second predefined parameter set to generate a plurality of second structured data files comprising a second DNA ID, and perform a second series of searches using progressively less conservative match criteria

Mixture-triggered threshold and limit parameter modifications for DNA IDs

modify one or more specified threshold, limit, and counting parameters when the at least one characteristic classifies sample data as a mixture

Visualization generation from structured DNA files

create a visualization of multiple structured data files for display on a display device

Match-score definition using specified metrics

define the match score using one or more metrics selected from random match probability and totals of matching alleles and loci

Coverage centers on determining characteristics including signal strength, generating structured DNA-related outputs, performing iterative searches with progressively less conservative match criteria and threshold scoring, and automatically iterating parameter-set values when searches do not meet threshold. The refinements further specify mixture-based parameter changes, visualization generation, and example match-score metrics.
