Abstract
Disclosed are compositions of matter, treatments and protocols useful for induction of T regulatory cells in response to inflammation, as well as inhibition of suicidal ideations and/or neuroinflammation. In some embodiments the invention teaches the administration of a therapeutic combination of ingredients comprising of minocycline, pterostilbene, Nigella sativa, sulforaphane, and epigallocatechin-3-gallate (EGCG) to a mammal undergoing upregulation of inflammatory mediators. In another embodiment, the invention teaches administration of said therapeutic combination to a mammal infected with said SARS-CoV-2 and/or possessing suicidal ideations. In some embodiments dosage of said therapeutic combination is based on inflammatory and/or immunological parameters observed in patients.
Core Innovation
The invention relates to nutraceutical-based therapy that induces inflammatory-condition-dependent T regulatory (Treg) cell generation and suppresses suicidal ideation and neuroinflammation. A subject in need of therapy receives a therapeutic combination comprising epigallocatechin-3-gallate (EGCG) or a green tea extract containing epigallocatechin-3-gallate, pterostilbene or a blueberry extract containing pterostilbene, thymoquinone or a Nigella sativa extract containing thymoquinone, sulforaphane or a broccoli extract containing sulforaphane, and minocycline.
The approach characterizes and leverages Treg cell phenotypes and functions, including FoxP3 expression, and suppressive activity of T regulatory cells against T cell receptor-stimulated CD3/CD28-activated T cells. Treg cell effects are associated with reduced neural inflammation and are connected to inflammation biomarker relationships such as enhanced TNF-alpha relative to an age-matched control, with TNF-alpha measured in cerebrospinal fluid or peripheral blood.
For reducing suicidal ideations, the invention selects a mammal in need of therapy and administers the same therapeutic combination. Suicidal ideation is assessed using named risk assessment tools, including the SAD PERSONS Scale, Manchester Self-Harm Rule (MSHR), Sodersjukhuset Self Harm Rule, and Beck Hopelessness Scale, and immunologic outcomes include changes in interleukin 10 (IL-10) and interleukin 6 (IL-6) production in peripheral blood mononuclear cells and microglial cells.
Claims Coverage
Two independent claims are identified. The claims cover stimulating production of T regulatory cells in a mammal and reducing suicidal ideations in a mammal, each using the same therapeutic combination.
Stimulating T regulatory cell production with a five-component therapeutic combination plus minocycline
A method of stimulating production of T regulatory cells in a mammal in need of therapy by selecting the mammal and administering a therapeutic combination comprising epigallocatechin-3-gallate or a green tea extract containing epigallocatechin-3-gallate, pterostilbene or a blueberry extract containing pterostilbene, thymoquinone or a Nigella sativa extract containing thymoquinone, sulforaphane or a broccoli extract containing sulforaphane, and minocycline.
Reducing suicidal ideations with the same five-component therapeutic combination plus minocycline
A method of reducing suicidal ideations by selecting a mammal in need of therapy and administering a therapeutic combination comprising epigallocatechin-3-gallate or a green tea extract containing epigallocatechin-3-gallate, pterostilbene or a blueberry extract containing pterostilbene, thymoquinone or a Nigella sativa extract containing thymoquinone, sulforaphane or a broccoli extract containing sulforaphane, and minocycline.
The claim coverage centers on administering a therapeutic combination containing EGCG or green tea extract, pterostilbene or blueberry extract, thymoquinone or Nigella sativa extract, sulforaphane or broccoli extract, and minocycline, either to stimulate T regulatory cell production or to reduce suicidal ideations. Dependent refinements include patient selection based on deficient Treg numbers or activity, quantifying FoxP3 and Treg suppressive activity, associating neuroinflammation with enhanced TNF-alpha, specifying TNF-alpha measurement location, assessing suicidal ideation using named tools, and immunologic outcome measures involving IL-10 or IL-6 production in peripheral blood mononuclear cells or microglial cells.
