Abstract

Compositions comprising one or more chelating agents and optionally zinc ion salts are used to inhibit the growth or biofilm formation in bacteria associated with personal care products such as ophthalmic, pedicure, manicure or podiatric solutions. The compositions of the present application can also comprise gelling agents, antimicrobials, antibiotic or a pH adjuster. The compositions may be in the form of a solution, a gel, a cream, a jelly, a powder, a paste, a lotion, soap and a cleaner.

Core Innovation

The invention provides antimicrobial-antibiofilm compositions comprising one or more chelating agents and optionally zinc ion salts, useful to inhibit growth and biofilm formation of bacteria associated with personal care products including ophthalmic, pedicure, manicure or podiatric solutions. These compositions may also include gelling agents, antimicrobials, antibiotics, or pH adjusters, and be formulated as solutions, gels, creams, jellies, powders, pastes, lotions, soaps, or cleaners.

The problem addressed is that microorganisms can adhere to personal care items such as contact lenses and containers, leading to contamination and infections. Existing multipurpose solutions are limited by the need to maintain ocular safety and comfort, restricting the types and amounts of antibacterial agents. There is a need for compositions with improved antibacterial and antibiofilm properties that maintain or increase biocidal efficacy without adversely affecting comfort or safety, and that can be used across a variety of personal care products.

The invention solves this need by providing compositions with two or more chelating agents combined with metal ion salts, particularly zinc salts, at effective concentrations to inhibit bacterial growth and biofilm formation. The invention further includes formulations that incorporate additional antimicrobial agents, surfactants, gelling agents, and personal care acceptable excipients to create effective anti-infective compositions suitable for various personal care product formulations and uses, including ophthalmic applications and treatments of pedicure, manicure, and podiatry instruments and solutions.

Claims Coverage

The patent includes one independent claim and several dependent claims detailing compositions and methods for inhibiting bacterial growth and biofilm formation in personal care products.

Composition comprising specific chelating agents and zinc salt at defined concentrations

A composition comprising water; one or more chelating agents selected from disodium or tetrasodium EDTA at at least about 62 mg/L; one or more chelating agents selected from sodium citrate and potassium citrate at about 3,000 mg/L; and a zinc salt selected from zinc chloride, zinc gluconate, zinc lactate, zinc citrate, zinc sulfate, and zinc acetate at about 100 mg/L.

Specific zinc salt selection

The zinc salt in the composition is zinc chloride or zinc citrate.

Inclusion of antimicrobial preservatives

The composition further includes antimicrobial preservatives selected from potassium sorbate, potassium benzoate, sodium benzoate, and benzoic acid.

Additional formulation ingredients

The composition may further comprise ingredients such as a buffer, stabilizing agent, gelling agent, surfactant, herbal, vitamin, mineral, extracellular matrix, antimicrobial, antibiotic, and pH adjuster.

Multiple formulation types

The composition can be prepared as a disinfecting solution, dip solution, lotion, cream, ointment, gel, or spray.

Use of delivery systems

The composition can be formulated with a liposome, nanoparticle, or suitable device delivery system.

Inclusion of anti-infective compounds

The composition can include anti-infective compounds such as glycoside hydrolases, alginate lyase, nisin, lactoferricin, serotransferrin, ovotransferrin, ovalbumin, ovomucoid, protamine sulfate, chlorhexidine, cetylpyridinium chloride, triclosan, silver sulfadiazine, benzalkonium chloride, hydrogen peroxide, citric acid, potassium citrate, 5-fluorouracil, cis-2-decenoic acid, DNase I, proteinase K, silver, gallium, bacteriocins, and antimicrobial peptides.

Inclusion of viscosity enhancers and other additives

The composition may further comprise viscosity enhancing agents, lubricants, surfactants, buffers, preservatives, and salts.

Form selection for application

The composition can be selected from solutions, gels, creams, jellies, powders, pastes, lotions, soaps, and cleaners.

Method of preventing or treating ophthalmic biofilm growth

Administering the composition to a user, including application to the eye to prevent or treat ophthalmic biofilm growth.

Method of application to other body parts

Administering the composition to a foot, fingernail, or toenail of a user as a method of treatment or prevention.

Personal care devices treated with the composition

Personal care devices treated or impregnated with the composition, including contact lenses and contact lens containers, and hand washing containers, scrubbers, and foot washing containers.

The claims cover compositions comprising combinations of specific chelating agents with zinc salts at defined concentrations, optionally including various antimicrobials, preservatives, and additives, formulated for multiple personal care product forms, and methods of using these compositions to inhibit bacterial growth and biofilm formation in personal care applications including ophthalmic use and treatment of personal care devices.

Stated Advantages

The composition enhances antibacterial and antibiofilm efficacy, improving inhibition of bacterial growth and biofilm formation compared to individual components alone.

Use of combinations of two or more chelating agents with zinc salts allows lower concentrations while maintaining effectiveness, reducing potential toxicity or irritation.

The compositions maintain or improve biocidal efficacy without adversely affecting comfort or safety, especially important for ocular applications.

The compositions have GRAS status and use ingredients that are safe as food and feed additives, supporting biocompatibility and safety.

Documented Applications

Use in ophthalmic personal care products including eye drops, eyewash solutions, contact lens cleaning, disinfecting, storing, and conditioning solutions, and contact lens containers.

Use in pedicure, manicure, and podiatry solutions, tools, and containers to prevent or treat microbial contamination and biofilm formation.

Treatment or impregnation of personal care products such as contact lenses, contact lens containers, hand washing containers, hand and foot scrubbers, and foot washing containers with the composition.
