Abstract

A method for treating a patient includes providing a delivery device capable of generating a magnetic field, placing the device above or below the skin, and directing the agent into the patient's skin via the device. The therapeutic agent is directed to a treatment site through the skin. Systems for directing agents are also included herein.

Core Innovation

The disclosed invention relates to directing a therapeutic agent into and/or through skin by magnetically assisted delivery. Magnetizable nanoparticles acted on by a magnetic field are used to direct the magnetizable nanoparticles into and through the skin, including ferro, ferri, paramagnetic, or super-paramagnetic nanoparticles.

The invention also includes a water repellant substance placed on the skin to contain the therapeutic agent, wherein the water repellant substance bounds the therapeutic agent. The magnetically driven nanoparticles are associated with the therapeutic payload, and may carry therapeutic payloads including therapeutic agents, drugs, proteins, or genes, including combinations.

A magnetic-field delivery configuration is described in terms of push versus pull magnet configurations and magnetic-field node concepts, including a minimum magnetic field and a concentration threshold behavior for delivery. The disclosed examples include magnetic-field-dependent delivery effects such as particle speed versus size/concentration and barrier penetration influenced by magnetic-field conditions and particle size/aggregation.

Claims Coverage

The partial content includes two independent claims. Each independent claim contains two main inventive elements directed to magnetically driving delivery into/through skin and bounding/containing the therapeutic agent with a water repellant substance on the skin.

Magnetically directed delivery into and through skin using magnetizable nanoparticles

A system comprising magnetizable nanoparticles acted on by a magnetic field to direct the magnetizable nanoparticles into and through the skin, wherein the magnetizable nanoparticles may be one or more of ferro, ferri, paramagnetic, or super-paramagnetic nanoparticles.

Water repellant substance bounding therapeutic agent on the skin

A water repellant substance placed on the skin to contain the therapeutic agent, wherein the water repellant substance bounds the therapeutic agent.

Magnetic-field-generating delivery device positioned relative to skin

A method providing a delivery device capable of generating a magnetic field; placing the device above, below or aside to a patient's skin.

Bounding water repellant substance and directing therapeutic agent to a treatment site

Placing a therapeutic agent within a bounding water repellant substance that is placed on the skin to contain the therapeutic agent; and directing the therapeutic agent into the patient's skin via the device, wherein the therapeutic agent is directed to a treatment site.

Across the independent claims, the core claim coverage centers on magnetically assisted delivery into/through skin using magnetizable nanoparticles and a magnetic-field-generating device, combined with containment of the therapeutic agent by a water repellant substance bounding/containing the therapeutic agent on the skin, with delivery directed to a treatment site.

Stated Advantages

Reduces need for needles/needle phobia.

Potential infectious disease transmission risk reduction.

Documented Applications

Magnetically assisted delivery into a rat paw model with delivery effects evaluated using Prussian blue staining.

Barrier penetration studies using agarose gel and tight-junction cell layers as a cell layer mimic of epidermis, including observation of magnetic-field-dependent delivery effects and effective penetration at different particle sizes.

Microfluidic measurements of particle speed versus particle size/concentration under magnetic-field conditions, including tracking of particle aggregation/chains.
