Abstract
Described herein are methods of treating wounds, the method comprising administering to a subject an effective amount of a composition comprising a soluble or derivatized chitosan wherein the soluble or derivatized chitosan when administered contacts the wound, thereby treating the wound.
Core Innovation
The invention relates to treating a wound in the eye of a subject by topically administering an aqueous pharmaceutical composition comprising dextrose, sterile water, and a soluble derivatized chitosan of formula (I). The derivatized chitosan has a repeating unit index n between 20 and 6000, and R1 substituents are independently selected from hydrogen and acetyl, with at least 25% H, at least 1% acetyl, and at least 2% formula (II) groups or guanidine-forming groups. The molecular weight of the derivatized chitosan is between about 10,000 and 350,000 Da.
The derivatized chitosan is further characterized by functionalization degree, degree of deacetylation, polydispersity index, and solubility in a pH range such as about 6.8 to about 7.4. The composition is described as suitable for eye wound treatment and as a soluble material in the aqueous pharmaceutical composition.
The disclosed treatment is associated with lower cumulative corneal scores 36 hours after administration compared to an eye wound treated with a composition consisting essentially of dextrose and sterile water. The content also states that the derivatized chitosan treatment reduces healing time, decreases inflammation, and reduces scarring in wound treatment contexts.
Claims Coverage
The independent claim covers topical treatment of an eye wound using an aqueous pharmaceutical composition comprising dextrose, sterile water, and a soluble derivatized chitosan of formula (I). Three inventive feature groups recur across the input items: the defined formula (I) structure, the compositional and molecular-weight limits, and the reported 36-hour corneal-score improvement versus a dextrose/sterile-water control.
Topical aqueous composition for eye wound treatment
A method of treating a wound in the eye of a subject by topically administering an effective amount of an aqueous pharmaceutical composition comprising dextrose, sterile water, and a soluble derivatized chitosan of formula (I).
Defined formula (I) derivatized chitosan
The soluble derivatized chitosan has n between 20 and 6000, and R1 substituents independently selected from hydrogen and acetyl, with at least 25% H, at least 1% acetyl, and at least 2% formula (II) groups or guanidine-forming groups.
Molecular weight and solubility constraints
The molecular weight of the derivatized chitosan is between about 10,000 and 350,000 Da, with additional disclosed parameters including functionalization degree, degree of deacetylation, polydispersity index, and solubility in a pH range such as about 6.8 to about 7.4.
Lower cumulative corneal scores at 36 hours versus dextrose and sterile water control
Thirty-six hours after administration, the eye wound has lower cumulative corneal scores compared to an eye wound treated with a composition consisting essentially of dextrose and sterile water.
The claims center on topical eye-wound treatment with an aqueous dextrose and sterile water composition containing a soluble derivatized chitosan of formula (I) under specified structural and molecular-weight constraints, together with lower cumulative corneal scores at 36 hours versus a dextrose and sterile water control.
Stated Advantages
Lower cumulative corneal scores 36 hours after administration compared to a composition consisting essentially of dextrose and sterile water.
Reduces healing time.
Decreases inflammation.
Reduces scarring.
