Abstract

The present disclosure provides an antibody, antigen binding fragment thereof, or an antibody conjugate thereof, useful in methods for treatment or diagnosis of conditions characterized by the expression of PTGFRN. The antibodies or antigen binding fragments thereof may also be used in imaging or detecting cells that express PTGFRN.

Core Innovation

The invention provides isolated antibodies or antigen binding fragments that specifically bind to PTGFRN. The disclosed antibodies are defined by heavy chain variable region CDR sequences and light chain variable region CDR sequences identified by specific SEQ ID NOs, including sets recited as SEQ ID NOs:5-7 for the heavy chain CDRs and SEQ ID NOs:8-10 for the light chain CDRs, as well as other heavy- and light-chain CDR sequence sets.

The antibody compositions are further characterized as antibodies or antigen binding fragments that can be internalized after binding and can be of different antibody or antigen binding fragment formats. The document also recites that the antibodies can be murine, human, humanized, or chimeric and can include IgG1 or IgG2 constant regions, with antibody fragment types and engineered single-domain or multivalent constructs described as permissible.

The document further discloses anti-PTGFRN antibody conjugates, including antibody-drug conjugates (ADCs), for treating and diagnosing PTGFRN-expressing cancers. PTGFRN is identified as the target of a monoclonal antibody 33B7/AG02, and the document describes PTGFRN cell-surface binding and internalization.

Claims Coverage

The identified independent claims are 1, 6, and 11. Across these independent claims, the core coverage is PTGFRN-specific isolated antibodies or antigen-binding fragments defined by three distinct heavy-chain and light-chain CDR sequence pairings, with internalization and format limitations as additional narrowing features.

PTGFRN-specific isolated antibody defined by CDR sequence sets (SEQ ID NOs 5-7 and 8-10)

An isolated antibody or antigen-binding fragment thereof comprising a heavy chain variable region comprising the CDR sequences SEQ ID NOs:5, 6 and 7 and a light chain variable region comprising CDR sequences SEQ ID NOs:8, 9 and 10, wherein the antibody specifically binds to PTGFRN.

PTGFRN-specific isolated antibody defined by CDR sequence sets (SEQ ID NOs 35-37 and 38-40)

An isolated antibody or antigen-binding fragment thereof comprising a heavy chain variable region comprising the CDR sequences SEQ ID NOs:35, 36 and 37 and a light chain variable region comprising CDR sequences SEQ ID NOs:38, 39 and 40, wherein the antibody specifically binds to PTGFRN.

PTGFRN-specific isolated antibody defined by CDR sequence sets (SEQ ID NOs 11-13 and 20-22)

An isolated antibody or antigen-binding fragment thereof comprising a heavy chain variable region comprising the CDR sequences SEQ ID NOs:11, 12 and 13 and a light chain variable region comprising CDR sequences SEQ ID NOs:20, 21 and 22, wherein the antibody specifically binds to PTGFRN.

The claim set centers on PTGFRN-specific isolated antibodies or antigen-binding fragments defined by three heavy-chain and light-chain CDR sequence combinations. Dependent claim language further adds internalization, specifies antibody provenance classes, IgG1 or IgG2 constant regions, and permits multiple antibody or fragment formats.

Stated Advantages

Provides selective anti-tumor activity of a 33B7-saporin ADC in vitro against PTGFRN+ cells.

Provides tumor growth inhibition in xenograft models.

Documented Applications
