Abstract

The present invention relates to the medical field, in particular to the treatment of neurological disorders. More specifically the present invention relates to a nanoparticle or nanoparticles' aggregate for use in prevention or treatment of a neurological disease or at least one symptom thereof in a subject without exposure of the nanoparticle or nanoparticles' aggregate to an electric field, and preferably without exposure thereof to any other external activation source, wherein the nanoparticle's or nanoparticles' aggregate's material is selected from a conductor material, a semiconductor material, an insulator material with a dielectric constant εijk equal to or above 200, and an insulator material with a dielectric constant εijk equal to or below 100. It further relates to compositions and kits comprising such nanoparticles and/or nanoparticles' aggregates as well as to uses thereof without exposure thereof to an electric field, and preferably without exposure thereof to any other external activation source.

Core Innovation

The invention describes nanoparticles or nanoparticle aggregates for preventing or treating a neurological disease selected from Parkinson's disease, Alzheimer's disease, epilepsy, obsessive compulsive disorder, autism spectrum disorder, depression disorder, dystonia, Tourette's syndrome, schizophrenia, stroke, aphasia, dementia, tinnitus, Huntington's disease, essential tremor, bipolar disorder, anxiety disorder, addiction disorder, and consciousness vegetative state, or at least one symptom thereof, by administering a composition comprising nanoparticles and/or nanoparticle aggregates to a subject. The method is based on administering nanoparticle material selected from conductor materials, organic sp2-carbon materials, semiconductor materials, and insulator materials, with specified physicochemical constraints for the nanoparticle or nanoparticle aggregate population.

The nanoparticle or nanoparticle aggregate material is further constrained by measurement-defined properties, including selection criteria based on a standard reduction potential E0 above 0.2 for conductor metals, contiguous sp2 hybridized carbon centers for organic materials, a semiconductor band gap Eg below 3.0 eV for specified semiconductor compositions, and an insulator dielectric constant εijk that is either equal to or above 200 or equal to or below 100, with particular listed insulator materials. The median largest core size of the nanoparticle or nanoparticle aggregate population is specified as at least 30 nm for relevant material categories, and the core is coated with a biocompatible coating that provides a neutral or a negative surface charge under specified solution conditions.

A key aspect is that the method does not include exposing the nanoparticles or nanoparticle aggregates to an electric field or any other external activation source. The nanoparticles or aggregates remain in the subject and are not used as carriers of therapeutic compound(s) or drug(s). The disclosure further identifies therapeutic relevance through neuron-network evidence using microelectrode array based neuronal network models of Parkinson's disease via MPP+ and Alzheimer's disease via amyloid-beta, where abnormal neuronal oscillation synchrony is described as normalized by nanoparticle administration.

Claims Coverage

The partial content includes two independent claims, one directed to a method and one directed to a composition or kit. Across the independent claims, the coverage is based on administering or providing at least two distinct nanoparticles or nanoparticle aggregates with tightly defined material classes and physicochemical conditions, including exclusion of electric field or external activation and exclusion of nanoparticle use as drug carriers for the method claim.

Nanoparticle-based method for neurological prevention or treatment without electric-field or external activation

A method for preventing or treating a neurological disease selected from Parkinson's disease, Alzheimer's disease, epilepsy, obsessive compulsive disorder, autism spectrum disorder, depression disorder, dystonia, Tourette's syndrome, schizophrenia, stroke, aphasia, dementia, tinnitus, Huntington's disease, essential tremor, bipolar disorder, anxiety disorder, addiction disorder, and consciousness vegetative state, or at least one symptom thereof in a subject by administering nanoparticle or nanoparticle aggregate, or a composition comprising nanoparticles and/or nanoparticle aggregates and a pharmaceutically acceptable support, where the nanoparticle material is selected from specified conductor, organic sp2-carbon, semiconductor (Eg below 3.0 eV), and insulator materials (defined εijk ranges), with defined measurement conditions and constraints on core size and biocompatible coating surface charge; and wherein the method does not include exposing the nanoparticles or nanoparticle aggregates to an electric field or any other external activation source, the nanoparticles or aggregates remain in the subject, and the nanoparticles or aggregates are not used as carriers of therapeutic compound(s) or drug(s).

Composition or kit of at least two distinct nanoparticles with specified material classes and physicochemical constraints

A composition or kit comprising at least two distinct nanoparticles and/or nanoparticle aggregates, where the material is selected from specified conductor metals (E0 above 0.2), organic materials with contiguous sp2 hybridized carbon centers, semiconductors with band gap Eg below 3.0 eV, and insulators with defined dielectric constant εijk ranges (with listed insulator materials), with defined measurement temperature and frequency; wherein the median largest core size is at least 30 nm for relevant material categories; and wherein the core is coated with a biocompatible coating providing a neutral or a negative surface charge under defined electrolyte concentration, nanoparticle concentration, and pH in water.

Across the independent claims, the core coverage centers on specified nanoparticle material classes with stringent constraints on core size, biocompatible coating producing neutral or negative surface charge under defined solution conditions, and defined measurement conditions; and in the method claim specifically excludes electric-field or external activation and excludes nanoparticle use as carriers of therapeutic compounds or drugs.
