Abstract
The invention provides a method to prepare a graft comprising a recellularized extracellular matrix of a mammalian liver, liver lobe or portion thereof, and a method of using the recellularized extracellular matrix of a mammalian liver, liver lobe or portion thereof.
Core Innovation
The invention relates to introducing to a mammal that has Type 1 diabetes or Type 2 diabetes a re-endothelialized extracellular matrix of a mammalian liver or portion thereof. The re-endothelialized extracellular matrix comprises a first population of engrafted mammalian cells and a second population of engrafted mammalian cells, and the first population comprises endothelial cells, stem cells, endothelial progenitor cells, or any combination thereof, while the second population comprises insulin producing cells.
The re-endothelialized extracellular matrix is produced such that the first population or the second population are xenogeneic to the extracellular matrix of the mammalian liver or a portion thereof. The introduction of the re-endothelialized extracellular matrix thereby at least partially treats the Type 1 diabetes or the Type 2 diabetes.
In one aspect, the insulin producing cells are engrafted within the re-endothelialized extracellular matrix and include islet cells and beta cells. In related embodiments, the decellularized extracellular matrix used as the basis for the re-endothelialized extracellular matrix comprises a substantially intact vascular network and is prepared from a liver, liver lobe, or portion, supporting re-endothelialization and recellularization with the specified cell populations.
Claims Coverage
The partial content identifies two independent claims. Across these claims, the core inventive content is the use of a re-endothelialized mammalian liver extracellular matrix containing engrafted endothelial/stem/endothelial progenitor cells and insulin producing cells, or a re-endothelialized decellularized liver extracellular matrix containing an engrafted insulin producing cell population, with xenogeneic relationships to the extracellular matrix, for at least partial treatment of Type 1 diabetes or Type 2 diabetes.
Re-endothelialized mammalian liver extracellular matrix with endothelial and insulin producing cell populations
Introducing to a mammal that has Type 1 diabetes or Type 2 diabetes a re-endothelialized extracellular matrix of a mammalian liver or portion thereof; the re-endothelialized extracellular matrix comprises a first population of engrafted mammalian cells and a second population of engrafted mammalian cells, wherein the first population comprises endothelial cells, stem cells, endothelial progenitor cells, or any combination thereof, and wherein the second population comprises insulin producing cells; and wherein the first population or the second population are xenogeneic to the extracellular matrix of the mammalian liver or a portion thereof, thereby at least partially treating the Type 1 diabetes or the Type 2 diabetes.
Re-endothelialized decellularized liver extracellular matrix with xenographic engrafted insulin producing cells
Introducing to a mammal that has Type 1 diabetes or Type 2 diabetes a re-endothelialized extracellular matrix of a decellularized extracellular matrix of a mammalian liver or a portion thereof; wherein the re-endothelialized extracellular matrix comprises a population of engrafted insulin producing cells; and wherein the engrafted insulin producing cells are xenographic to the decellularized extracellular matrix of the mammalian liver or portion thereof, thereby at least partially treating the Type 1 diabetes or the Type 2 diabetes.
Both independent claims cover treating Type 1 diabetes or Type 2 diabetes by introducing a re-endothelialized extracellular matrix derived from mammalian liver, or a portion thereof, where the matrix contains engrafted insulin producing cells and at least one specified cell population is xenogeneic or xenographic relative to the liver extracellular matrix. One independent claim additionally specifies an engrafted first population comprising endothelial cells, stem cells, endothelial progenitor cells, or combinations thereof.
Stated Advantages
At least partial treatment of Type 1 diabetes or Type 2 diabetes.
Documented Applications
Treating a mammal with Type 1 diabetes or Type 2 diabetes by introducing a re-endothelialized mammalian liver extracellular matrix containing engrafted endothelial/stem/endothelial progenitor cells and insulin producing cells, or an engrafted insulin producing cell population.
