Abstract

Bead arrays suitable for analysis by mass spectrometry are disclosed. In an embodiment, a bead array includes multiple reactive sites, each of the reactive sites being capable of binding multiple distinct target analytes.

Core Innovation

A bead array system is adapted for use in analyzing a sample to identify at least one target from among a set of potential targets with known identities. The system includes a bead array having multiple reactive sites, where a first reactive site has a first bead with multiple copies of a first capture agent that specifically recognizes a first epitope and is configured to specifically bind, but has not yet bound, any of a plurality of first reactive site potential targets containing the first epitope.

A second reactive site includes a second bead with multiple copies of a second capture agent distinct from the first capture agent. Each copy of the second capture agent specifically recognizes a second epitope and is configured to specifically bind, but has not yet bound, any of at least one second reactive site potential target containing the second epitope, thereby enabling discrimination among targets that can bind to different capture-agent/epitope combinations.

The system further includes a decoding table in an electronic medium that determines the identity of each target that will bind to the first capture agent or the second capture agent. The decoding table contains information collected prior to contacting the sample with the first and second reactive sites, including molecular-weight-derived values for the potential targets and expected abundances of the first and the second target in the sample.

Claims Coverage

Only one independent claim is provided in the partial content. It defines a bead array system in combination with an electronic decoding table, using two distinct reactive sites and pre-collected molecular-weight-derived values plus expected abundances to determine which known targets will bind each reactive site.

Two reactive sites with epitope-specific capture agents and distinct targets

A bead array having a first reactive site with a first bead and multiple copies of a first capture agent that specifically recognizes a first epitope and binds potential targets containing the first epitope, and a second reactive site with a second bead and multiple copies of a second capture agent distinct from the first capture agent that specifically recognizes a second epitope and binds at least one second reactive site potential target containing the second epitope.

Pre-collected molecular-weight-derived decoding table with expected abundances

An electronic decoding table configured to determine the identity of each target that will bind to the first capture agent or the second capture agent, where the table contains information collected prior to contacting the sample with the reactive sites, including molecular-weight-derived values for the potential targets and expected abundances of the first and the second target in the sample.

The independent claim coverage centers on two distinct reactive sites on a bead array, each defined by epitope-specific capture agents, together with an electronic decoding table containing pre-collected molecular-weight-derived values and expected abundances to determine which known targets bind each capture agent.
