Abstract
The present invention provides a process for the preparation of a Crassocephalum crepidioides extract, and the extract prepared thereby. The present invention further relates to a pharmaceutical composition/combination comprising the Crassocephalum crepidioides extract. The use of the extract and the composition/combination comprising the extract in the prevention or treatment of cancer is also provided.
Core Innovation
The invention relates to a process for preparing an extract of Crassocephalum crepidioides. The process includes contacting a Crassocephalum crepidioides plant or part thereof with 70% ethanol to obtain a suspension comprising a solid portion and a liquid portion, and separating the solid portion to obtain a crude extract. The crude extract is diluted to obtain a 60% ethanol extract.
The 60% ethanol extract is mixed with macroporous styrene-divinyl benzene resins and poured into a column. The column is washed with ethanol, and the column is eluted with a solvent of ethanol/ethyl acetate or ethyl acetate while collecting eluted fractions at different time intervals. Each eluted fraction is analyzed to determine which fraction is enriched in 1,2-di-O-α-linolenoyl-3-O-β-galactopyranosyl-sn-glycerol (dLGG) and contains phytol.
The fraction enriched in dLGG and containing phytol is recovered to obtain the Crassocephalum crepidioides extract. The document further describes characterization of the extract using HPLC and describes pharmaceutical compositions and combination regimens involving the Crassocephalum crepidioides extract for cancer prevention and treatment.
Claims Coverage
The document provides one independent claim. The claim coverage centers on an ethanol-based extraction and macroporous styrene-divinyl benzene resin chromatography workflow that yields a Crassocephalum crepidioides extract enriched in dLGG and containing phytol.
Ethanol contacting and crude extract preparation
Contacting a Crassocephalum crepidioides plant or part thereof with 70% ethanol to obtain a suspension comprising a solid portion and a liquid portion, separating the solid portion from the liquid portion, and collecting the liquid portion to obtain a crude extract.
Dilution to a 60% ethanol extract
Diluting the crude extract to a 60% ethanol extract.
Purification using macroporous styrene-divinyl benzene resin column
Mixing the 60% ethanol extract with macroporous styrene-divinyl benzene resins, pouring the mixture into a column, and washing the column with ethanol.
Elution and fraction selection enriched in dLGG and phytol
Eluting the column with ethanol/ethyl acetate or ethyl acetate, collecting eluted fractions at different time intervals, analyzing each fraction, and recovering the fraction enriched in 1,2-di-O-α-linolenoyl-3-O-β-galactopyranosyl-sn-glycerol (dLGG) and containing phytol to obtain the Crassocephalum crepidioides extract.
Overall, the independent claim protects an ethanol extraction combined with macroporous styrene-divinyl benzene resin chromatography, where eluted fractions are analyzed and selected to recover a Crassocephalum crepidioides extract enriched in dLGG and containing phytol.
Stated Advantages
