Abstract

The present disclosure relates to compositions comprising a hydrogel particle with optical properties substantially similar to the optical properties of a target cell, and methods for their use.

Core Innovation

The invention uses hydrogel particles whose optical scatter property is at least 70% similar to that of a target cell type to calibrate a cytometric device. The optical scatter property is provided by a co-monomer, a chemical side-group, an encapsulated material, colloidal silica, or a ratio of acrylamide to bi-acrylamide. Particle composition and incorporated materials tune forward scatter and side scatter, including adjustment of side scatter without substantially changing forward scatter.

The calibrated cytometric device is used to analyze the target cell population after a physical property of the hydrogel particles is measured. The described approach provides synthetic, tunable calibrants as alternatives to purified cell lines, which are described as variable, and polystyrene beads. The partial content presents this approach for cell-like scatter calibration and subsequent cell-population analysis.

Claims Coverage

The claims identify three inventive features directed to calibrating a cytometric device with hydrogel particles and analyzing a target cell population using the calibrated device.

Hydrogel particles with target-cell-like optical scatter

The claim specifies hydrogel particles having an optical scatter property at least 70% similar to that of at least one target cell type associated with a target cell population, wherein the property is provided by a co-monomer, a chemical side-group, an encapsulated material, colloidal silica, or a ratio of acrylamide to bi-acrylamide.

Measurement of a particle physical property for calibration

The claim includes measuring a physical property of the hydrogel particles using the cytometric device as part of calibrating that device.

Analysis of the target cell population

The claim uses the calibrated cytometric device to analyze the target cell population.

Claim 1 combines target-cell-like optical scatter in hydrogel particles, measurement of a particle physical property for device calibration, and subsequent analysis of the target cell population with the calibrated device.

Stated Advantages

The approach provides synthetic, tunable calibrants as alternatives to variable purified cell lines and polystyrene beads.

Documented Applications

Calibration of a cytometric device and subsequent analysis of a target cell population using the calibrated device.
