Abstract
Preparations of glycan therapeutics, pharmaceutical compositions, dietary supplements and medical foods thereof are provided, and methods of using said gycan therapeutics, e.g. in cancer therapy.
Core Innovation
The invention provides a method of treating cancer in a subject by administering an effective amount of a composition comprising a glycan therapeutic preparation. The glycan therapeutic preparation comprises branched glycans comprising glucose, galactose, arabinose, mannose, fructose, xylose, fucose, or rhamnose glycan units, and the average degree of branching (DB) is at least 0.01.
At least 50% of the glycans have a degree of polymerization (DP) of at least 3 and less than 30 glycan units, and the ratio of alpha- to beta-glycosidic bonds present in the glycans is between about 1:1 and about 5:1. In some embodiments, the composition has a final water solubility limit in water of at least about 60 Brix at 23°C.
The method includes administering the composition in combination with an anti-cancer drug or anti-cancer therapy. The compositions are also described as pharmaceutical compositions, medical foods, dietary supplements, unit dosage forms, and kit concepts, with optional prebiotics, probiotic bacteria, and micronutrients.
Claims Coverage
The relevant independent claim is directed to treating cancer using a glycan therapeutic preparation with defined branched glycan structural parameters, together with an anti-cancer drug or anti-cancer therapy. The consolidated content identifies three core inventive features, with related dependent features adding solubility and outcome limitations.
Branched glycan units and minimum degree of branching for a cancer treatment composition
A composition comprising a glycan therapeutic preparation, where the glycan therapeutic preparation comprises branched glycans comprising glucose, galactose, arabinose, mannose, fructose, xylose, fucose, or rhamnose glycan units, and the average degree of branching (DB) of the branched glycans is at least 0.01.
Degree of polymerization range and alpha to beta glycosidic bond ratio
At least 50% of the glycans have a degree of polymerization (DP) of at least 3 and less than 30 glycan units, and the ratio of alpha- to beta-glycosidic bonds present in the glycans is between about 1:1 and about 5:1.
Combination administration with an anti-cancer drug or anti-cancer therapy
The composition is administered in combination with an anti-cancer drug or anti-cancer therapy as part of a method of treating cancer in a subject, using an effective amount of the composition.
Coverage centers on administering a glycan therapeutic preparation with specified branched glycan unit types, minimum degree of branching, defined DP distribution, and an alpha to beta glycosidic bond ratio, together with an anti-cancer drug or anti-cancer therapy. Optional content includes a final water solubility limit of at least about 60 Brix at 23°C and an increased white blood cell count compared to a control subject.
Stated Advantages
Enabling subject selection and monitoring based on microbiota or metabolite profiles.
Reducing therapy side effects or toxicity.
The partial content describes expected immune cell changes and increased immune surveillance in connection with glycan therapeutics used for cancer and immune/nutritional imbalances, including changes involving Th17/Th1 and cytotoxic T/NK activity.
The partial content describes shifts in the microbiome, including beneficial taxa and tumor-infiltrating bacteria, and effects of microbial metabolites such as SCFAs and bile acids.
The partial content describes administration concepts using responder/non-responder criteria and monitoring via microbiome/proteome/metabolome/transcriptome/proteome profiling.
After administering the composition, the subject’s white blood cell count is increased compared to a control subject.
Documented Applications
