Abstract

In a composition and a microneedle for a localized obesity treatment containing the same in accordance with the present disclosure, the composition in accordance with the present disclosure contains a gelatin, decomposes a fat in a body, and inhibits lipogenesis and fat deposition in the body.

Core Innovation

The patent discloses methods for treating an individual having localized obesity by administering a composition comprising animal-derived gelatin at a location where the individual has localized obesity. The composition is administered by injection with a hypodermic needle or with a microneedle to skin tissue below the skin stratum corneum. The composition reduces adipocyte size in adipose tissue at the location where the individual has localized obesity.

The microneedle system is configured for delivery into skin tissue below the skin stratum corneum, where the microneedle tip reaches a target region and enables the localized delivery of the animal-derived gelatin. The localized delivery is associated with reducing adipocyte size and with inhibiting lipogenesis and fat deposition, together with local adipose transcriptional regulators such as FASN, SREBP-1c, and PPARG.

In embodiments, gelatin is cross-linked, including cross-linking with genipin, to enable sustained release of the gelatin over at least about three days. The disclosed compositions and delivery approach are supported by in vitro and in vivo rat data showing anti-lipogenic, lipolytic, and adipose-deposition-inhibiting effects, including sustained effects with genipin-crosslinked gelatin microneedles and fat-related effects with injectable gelatin formulations.

Claims Coverage

The independent claim in the provided material is directed to a method of treating localized obesity using an animal-derived gelatin composition delivered below the skin stratum corneum by hypodermic needle injection or by microneedle administration. The coverage centers on three main inventive features: targeted delivery below the stratum corneum, reduction of adipocyte size, and optional genipin cross-linking.

Localized obesity treatment with animal-derived gelatin at the obesity location

A method for treating an individual having localized obesity comprising administering a composition comprising animal-derived gelatin to the individual at a location where the individual has localized obesity, wherein the composition reduces adipocyte size in adipose tissue at the location where the individual has localized obesity.

Administration below the skin stratum corneum by injection or microneedle

The composition is administered by injection with a hypodermic needle or with a microneedle to skin tissue below the skin stratum corneum.

Microneedle delivery enabling localized reduction of adipose function

A method in which administration is performed with a microneedle so that the composition is administered at skin tissue below the skin stratum corneum, where the microneedle tip decompresses to release the composition that locally reduces adipocyte size and locally inhibits lipogenesis and fat deposition.

Genipin cross-linked gelatin for sustained release

In embodiments, the composition comprises cross-linked gelatin that is cross-linked using genipin.

Stratum corneum-constrained microneedle tip geometry

A microneedle having a tip portion whose length is equal to or greater than the thickness of a skin stratum corneum of the individual.

Across the independent claim and the provided refinements, the inventive coverage is directed to delivering an animal-derived gelatin composition to the localized obesity site below the skin stratum corneum, via hypodermic needle injection or microneedle, to reduce adipocyte size, with dependent features emphasizing microneedle-enabled localized inhibition of lipogenesis and fat deposition and optional genipin cross-linking.

Stated Advantages

Reduces adipocyte size in adipose tissue at the location where the individual has localized obesity.

Locally inhibits lipogenesis and fat deposition.

Provides sustained release with genipin-crosslinked gelatin microneedles over at least about three days.

Documented Applications

Treating an individual having localized obesity, including localized obesity and related cellulite treatment contexts described in the disclosure.

Localized adipose tissue targeting at a skin site below the skin stratum corneum using a microneedle or hypodermic needle administration of animal-derived gelatin.

In vivo rat applications supporting anti-lipogenic, lipolytic, and adipose-deposition-inhibiting effects, including sustained effects with genipin-crosslinked gelatin microneedles and abdominal/visceral fat effects with injectable gelatin formulations.
