Abstract

The present invention provides novel compositions comprising multipotent cells or microvascular tissue, wherein the cells or tissue has been sterilized and/or treated to inactivated viruses, and related methods of using these compositions to treat or prevent tissue injury or disease in an allogeneic subject.

Core Innovation

The invention provides a method of preparing a sterilized composition that includes isolated multipotent cells. The method includes dissociating a tissue sample obtained from a donor mammal to release a plurality of multipotent cells and separating the released multipotent cells from one or more other tissue components to produce a composition comprising isolated multipotent cells. The resulting composition is sterilized while retaining angiogenic or anti-inflammatory activity, and the tissue is adipose-derived tissue, bone marrow, bone, muscle tissue, umbilical cord tissue, or amniotic tissue.

The invention further provides a method of preparing a sterilized composition comprising processed microvascular tissue. The method includes dissociating a microvascular tissue sample obtained from a donor mammal to produce a composition comprising dissociated microvascular tissue, removing one or more tissue components from the composition, and sterilizing the composition resulting after removal. The composition retains angiogenic or anti-inflammatory activity, and the microvascular tissue is limited to adipose-derived tissue, bone marrow, bone, muscle tissue, umbilical cord tissue, or amniotic tissue.

Additional processing steps associated with sterilization and preservation are described while maintaining retained angiogenic or anti-inflammatory activity. The additional processing includes drying, lyophilizing, or cryopreserving, and may include inactivating a virus in the composition. The dissociation aspects are also described as using protease(s) that do not include collagenase, and released multipotent cells are not cultured.

Claims Coverage

The patent identifies two independent claims that cover preparing sterilized, composition-forming products that retain angiogenic or anti-inflammatory activity, one for isolated multipotent cells and one for processed microvascular tissue. Each independent claim is anchored by a processing workflow and tissue source limitations.

Sterilized composition of isolated multipotent cells retaining angiogenic or anti-inflammatory activity

A method comprising dissociating a donor-mammal tissue sample to release a plurality of multipotent cells, separating the released multipotent cells from one or more other tissue components to produce a composition comprising isolated multipotent cells, and sterilizing the resulting composition, wherein the composition retains angiogenic or anti-inflammatory activity and the tissue is adipose-derived tissue, bone marrow, bone, muscle tissue, umbilical cord tissue, or amniotic tissue.

Sterilized composition of processed microvascular tissue retaining angiogenic or anti-inflammatory activity

A method comprising dissociating a donor-mammal microvascular tissue sample to produce a composition comprising dissociated microvascular tissue, removing one or more tissue components from the composition produced according to dissociation, and sterilizing the composition following removal, wherein the composition retains angiogenic or anti-inflammatory activity and the tissue is adipose-derived tissue, bone marrow, bone, muscle tissue, umbilical cord tissue, or amniotic tissue.

Collectively, the independent claims define sterilized compositions derived from specified donor-mammal tissue sources, where sterilization is performed on either isolated multipotent cells or processed microvascular tissue while retaining angiogenic or anti-inflammatory activity.

Stated Advantages

The sterilized compositions retain angiogenic or anti-inflammatory activity.
