Abstract
Provided herein include methods and compositions for treating diseases or conditions. In some embodiments provided are methods for treating one or more diseases or conditions selected from the group consisting of hypertension, heart failure, dyspnea, and sleep apnea. In certain embodiments provided are methods that include administering a compound of formula (I) as disclosed herein. In some embodiments provided are methods that include administering a P2X3 and/or a P2X2/3 receptor antagonist.
Core Innovation
The disclosure relates to therapeutic methods for treating sleep apnea in a subject in need thereof. The methods include administering an effective amount of a compound of formula (I) or a pharmaceutically acceptable salt thereof, and further include a specified compound or a pharmaceutically acceptable salt of a specified substance.
Formula (I) is defined by substituent parameters R1 and R2, with exemplary formula (I) compounds listed in Table 1. The document characterizes sleep apnea by central sleep apnea, obstructive sleep apnea, or mixed sleep apnea, and by apneic event frequency thresholds including more than about 5 apneic events per hour and more than about 15 apneic events per hour.
The disclosure also uses language directed to carotid body tonicity/activity, carotid glomus, and modulation of carotid chemosensory afferent discharge. In some embodiments, it includes a P2X3 receptor antagonist and/or a P2X2/3 receptor antagonist, and relates to ATP-gated purinergic signaling linked to carotid body hypertonicity and hypersensitivity.
Claims Coverage
The claim coverage includes three independent claims focused on methods of treating sleep apnea by administering an effective amount of a compound of formula (I), a specified compound, or a pharmaceutically acceptable salt. Across the independent claims, the inventive features center on the sleep apnea treatment framework and specific compound or salt selection, with dependent refinements to sleep apnea subtype and apneic-event frequency thresholds.
Administering an effective amount of compound of formula (I) for sleep apnea
A method of treating sleep apnea in a subject in need thereof comprising administering an effective amount of a compound of formula (I) or a pharmaceutically acceptable salt thereof, with R1 and R2 defined by the listed substituent options.
Administering an effective amount of a specified compound for sleep apnea
A method of treating sleep apnea in a subject in need thereof comprising administering to the subject an effective amount of the specified compound or a pharmaceutically acceptable salt thereof.
Administering an effective amount of a pharmaceutically acceptable salt for sleep apnea
A method of treating sleep apnea in a subject in need thereof comprising administering to the subject an effective amount of a pharmaceutically acceptable salt of a specified substance.
Across the independent claims, the core coverage is a sleep apnea treatment method in which an effective amount is administered, with the identity of the administered agent defined as either a compound of formula (I), a specific compound, or a pharmaceutically acceptable salt. Dependent claim refinements narrow the treatment to central or obstructive sleep apnea and apply apneic-event frequency thresholds.
Stated Advantages
Reducing carotid sinus nerve activity and excitatory responses to hypoxia/ATP in ex vivo mouse preparations.
Attenuating carotid body tonic activity, sympathetic discharge, and blood pressure in spontaneously hypertensive rats.
Modulating ATP-gated purinergic signaling associated with carotid body hypertonicity and hypersensitivity for therapeutic benefit in sleep apnea and related sympathetic overdrive diseases.
Documented Applications
Treating sleep apnea in a subject in need thereof by administering compounds of Formula (I), a specified compound, or pharmaceutically acceptable salts.
Treating diseases associated with heightened sympathetic activity and carotid body hypertonicity and/or hypersensitivity, including hypertension, heart failure, dyspnea, and chronic sleep apnea.
Treating sleep apnea in a subject in need thereof, including central sleep apnea, obstructive sleep apnea, or mixed sleep apnea, and characterizing severity using apneic events per hour.
