Abstract

A fluorocarbon emulsion in water for use in fractionated radiotherapy and chemotherapy, wherein said fluorocarbon comprises between 4 and 8 carbon atoms.

Core Innovation

The invention provides a method of sensitizing a mammal to multi-fraction radiotherapy or chemotherapy by administering a water emulsion of perfluoropentane. The regimen includes a phospholipid, a viscosity modifier, and a fluorosurfactant, together with about 1% to 5% w/vol of perfluoropentane, and a perfluoropentane emulsion dosage in the range from about 0.05 cc/kg to about 0.3 cc/kg.

The method synchronizes oxygen therapy with the cancer treatment schedule. The perfluoropentane emulsion is administered concomitantly or up to 120 minutes prior to each fraction of radiation therapy, or concomitantly with chemotherapy, and carbogen or supplemental oxygen is simultaneously administered to the mammal.

The invention addresses tumor hypoxia limiting radiosensitization. It uses the combination of carbogen or supplemental oxygen with the perfluoropentane emulsion to increase tumor oxygenation during treatment, with mention of comparable effects between carbogen and oxygen, monitoring of tumor oxygenation and tumor response, and gene-expression normalization via reduced hypoxia-related gene expression after DDFPe treatment.

Claims Coverage

The independent claim is clm-00001 and includes 4 inventive features.

Multi-fraction sensitization with perfluoropentane water emulsion and oxygen therapy

Sensitizing a mammal to multi-fraction radiotherapy or chemotherapy by administering a water emulsion of perfluoropentane and simultaneously administering carbogen or supplemental oxygen.

Concomitant or up-to-120-minute timing relative to radiation fractions or chemotherapy

Administering the perfluoropentane emulsion concomitantly or up to 120 minutes prior to each fraction of radiation therapy, or administering it concomitantly with chemotherapy.

Specified emulsion components and perfluoropentane content

The perfluoropentane emulsion comprises a phospholipid, a viscosity modifier, a fluorosurfactant, and about 1% to 5% w/vol of perfluoropentane.

Dose range for perfluoropentane emulsion

Administering the perfluoropentane emulsion at a dosage in the range from about 0.05 cc/kg to about 0.3 cc/kg.

Sensitization is achieved by synchronizing a specified perfluoropentane water emulsion with simultaneous carbogen or supplemental oxygen, using a concomitant or up-to-120-minute timing relationship to radiation fractions or chemotherapy.

Stated Advantages

Enhances tumor oxygenation during treatment.

Increases radiosensitization by addressing limitations imposed by tumor hypoxia.

Comparable effects are mentioned between carbogen and oxygen.

Documented Applications

Sensitizing mammals to multi-fraction radiotherapy and/or chemotherapy using carbogen or supplemental oxygen together with a perfluoropentane emulsion, with monitoring of tumor oxygenation and tumor response.

Tumor oxygenation monitoring and tumor response assessment are described together with gene-expression normalization via reduced hypoxia-related gene expression after DDFPe treatment.
