Abstract

Microarray compositions suitable for analysis by one or several spectrographic methods are disclosed. In an embodiment, a microarray composition includes a three-dimensional solid support and a plurality of reactive microbeads positioned on the solid support in spatially distinct and addressable locations.

Core Innovation

A composite microarray is provided with a solid support comprising an analytical site and a reactive site, where the reactive site is distinct from the analytical site. The reactive site includes a bead and an active agent associated with the bead, where the active agent is configured to bind a compound. The solid support is configured to provide a fluidic connection between the reactive site and the analytical site, and the dimensions of the analytical site do not exceed 1 mm.

The composite microarray supports migration of bound compound from the bead at the reactive site toward the analytical site such that at least a portion of the bound compound migrates from the bead to the analytical site. In the documented compositions, the migration results in localization of released analytes as uniform spots for spectrographic readout.

The disclosed platform is associated with analysis modalities including mass spectrometry and optical spectroscopy, including mass spectrometry performed as matrix-assisted laser desorption ionization (MALDI) and fluorescence spectroscopy. Documented configurations include ordered microwell layouts and fluidically coupled analytical sites, together with encoding and decoding strategies such as positional encoding, optical encoding, mass-tag encoding, and encoder-less modes.

Claims Coverage

The independent claim set identifies composite microarray platforms and a method of preparing a sample for analysis. Across these independent claims, the core inventive features are a distinct analytical site and reactive site on a solid support, a bead-associated active agent configured to bind a compound, an analytical site dimension limitation not exceeding 1 mm, and a fluidic connection enabling migration from the reactive site to the analytical site.

Distinct analytical and reactive sites on a solid support with a fluidic connection

A composite microarray comprising a solid support with an analytical site and a reactive site that is distinct from the analytical site, where the solid support is configured to provide a fluidic connection between the reactive site and the analytical site.

Bead-associated active agent configured to bind a compound

The reactive site comprises a bead and an active agent associated with the bead, where the active agent is configured to bind a compound.

Analytical site dimension does not exceed 1 mm

Dimensions of the analytical site do not exceed 1 mm.

Migration of bound compound from the bead to the analytical site

Effectuating migration of at least a portion of the bound compound from the bead of the reactive site to the analytical site, wherein the reactive site is distinct from the analytical site.

The independent claims consistently require a distinct reactive site and analytical site on a solid support with a fluidic connection, use a bead-associated active agent to bind a compound, limit analytical site dimensions to not exceed 1 mm, and rely on migration from the reactive-site bead to the analytical site as the key mechanism for analytical localization or readout.

Stated Advantages

The migration results in localization of released analytes as uniform spots for spectrographic readout.

Documented Applications

Analysis using mass spectrometry, including matrix-assisted laser desorption ionization (MALDI).

Analysis using optical spectroscopy, including fluorescence spectroscopy.

Positional encoding, optical encoding, mass-tag encoding, and encoder-less modes.
