Abstract
The present invention relates to a pharmaceutical composition for preventing, treating or delaying an Alzheimer's disease (AD) or dementia including a G protein-coupled receptor19 (GPCR19) agonist or its pharmaceutically acceptable salt as an active ingredient and a food composition for preventing, treating or delaying an Alzheimer's disease or dementia. The GPCR19 agonist according to the present invention has an effect of improving cognitive and behavioral disorders without harming the health of objects when administrated to the objects and has an effect of preventing the Alzheimer's disease or dementia or delaying or treating the progression of the disease in the objects with the disease by suppressing apoptosis of the brain tissue, enhancing immunity, and reducing formation of an amyloid β (Aβ) plague.
Core Innovation
The disclosed invention relates to a pharmaceutical and food composition for preventing, treating, or delaying Alzheimer’s disease or dementia. The composition includes a GPCR19 agonist, wherein the GPCR19 agonist is sodium taurodeoxycholate, including derivatives and/or salts, as an active ingredient, and is formulated to deliver a pharmaceutically effective amount to an individual having the disease.
The invention is directed to a method for treating an Alzheimer’s-related disease or dementia-related disease. The method comprises administering the pharmaceutical composition containing a GPCR19 agonist, specifically sodium taurodeoxycholate, to an individual having a disease selected from Alzheimer’s disease-related conditions and dementia-related conditions, including degenerative dementia and senile dementia.
The disclosure further describes disease-related effects associated with GPCR19 agonist use, including suppression of brain tissue apoptosis, enhanced immunity, and reduction of amyloid-β plaque and amyloid-associated inflammation, together with cognitive/behavioral improvement. Example work in a 5×FAD mouse model is described to support these effects through weight safety after repeated dosing and improved spatial learning ability.
Claims Coverage
The partial claim set includes one independent claim and multiple dependent claims, centered on administering a GPCR19 agonist where the GPCR19 agonist is sodium taurodeoxycholate, with dependent refinements specifying administration routes, daily dose ranges, and a weekly dosing frequency.
Treating Alzheimer’s-related and dementia-related diseases with sodium taurodeoxycholate as a GPCR19 agonist
A method for treating an Alzheimer’s-related disease or a dementia-related disease by administering a pharmaceutical composition containing a pharmaceutically effective amount of a GPCR19 agonist or a pharmaceutically acceptable salt thereof to an individual having the disease, wherein the GPCR19 agonist is sodium taurodeoxycholate.
Multiple administration/formulation routes for the pharmaceutical composition
The method includes administering the pharmaceutical composition formulated for administration by an oral, rectal, intravenous, intramuscular, intraperitoneal, subcutaneous, intranasal, or percutaneous route.
Daily dose range (0.1 mg/kg/day to 1000 mg/kg/day)
The method includes administering the GPCR19 agonist or a pharmaceutically acceptable salt thereof at a daily dose ranging from 0.1 mg/kg/day to 1000 mg/kg/day.
Daily dose range (1 mg/kg/day to 500 mg/kg/day)
The method includes administering the GPCR19 agonist or a pharmaceutically acceptable salt thereof at a daily dose of 1 mg/kg/day to 500 mg/kg/day.
Daily dose range (10 mg/kg/day to 100 mg/kg/day)
The method includes administering the GPCR19 agonist or a pharmaceutically acceptable salt thereof at a daily dose of 10 mg/kg/day to 100 mg/kg/day.
Weekly dosing frequency (1 to 3 times per week)
The method includes administering the GPCR19 agonist or a pharmaceutically acceptable salt thereof 1 to 3 times per week.
Overall claim coverage is centered on treating Alzheimer’s-related disease or dementia-related disease by administering a pharmaceutical composition containing a pharmaceutically effective amount of a GPCR19 agonist where the GPCR19 agonist is sodium taurodeoxycholate, with dependent refinements specifying administration routes, daily dose ranges, and a weekly dosing frequency.
